Technical Summary
The Mithen research group at IFR have developed a series of near-isogenic broccoli genotypes with varying levels of 4-methylsulphinylbutyl glucosinolate (glucoraphanin) due to allelic variation at the Myb28 locus. These broccoli genotypes have been processed into soups and are being used as part of a long term dietary intervention study. This project is concerned with a detailed analyses of sulphur-metabolites found in human plasma and urine following consumption of these soups. It involves a three arm doubled blinded cross over study with ten volunteers. Each arm consists of consuming a single 300 ml portion of soup made from one of three broccoli genotypes of different Myb28 genotypes. Blood and urine samples are collected over the 48 hours following consumption for analysis of a range of sulphur-containing metabolites derived from glucoraphanin. The human study is complemented with exploring the biological activity of these S metabolites particularly regarding their ability to enhance expression of genes of the pentose phosphate pathway in human liver cells.
