Technical Summary

The objectives of the project are: 1) to genetically and functionally characterise the cj1321-26 glycosylation island, a range of mutants and complemented mutants will be constructed and evaluated in several in vitro assays relevant to cell surface properties; 2) to ascertain if selective pressure is required to maintain the cj1321-26 glycosylation island in the avian host, the rate of spontaneous loss of this locus will be compared in vitro and in vivo; 3) to resolve the extent and structure of glycosylation on the NCTC11168H flagellin, NMR and mass spectroscopy will be applied to characterise the parent NCTC11168H strain and mutants generated in 1); 4) to confirm that the cj1321-26 glycosylation island is directly important in avian niche adaptation, NCTC11168H, mutants and complemented mutants generated in 1) will be tested for their ability to colonise and survive in chickens and induce immune responses.
