Role of fatty acid chain length in energy balance and adaptive thermogenesis

Abstract
Food contains different types of fat. There are some suggestions that not all the fats are equal and that there is good and bad fat with respect to their effects in the body. Not too much is known about what fat normally does in the cells of the body. We know that fat can be stored, burned, been sent from one organ to another within the body and more importantly fat are important 'bricks' to build up the different components of the cells. Interestingly not all the fats have the same size, the length of some fats is longer than others and we think that the length of the fat species may be an important determinant whether fat is stored or burn. The best organ to study this is a special type of organ known as brown fat present in children and small animals. This organ can do both, making fat and burning fat so it is the ideal organ to investigate the effect of fat length on these two processes. To study fat length we will investigate the effect of lack and/or excess of Elovl6, a molecule that elongates fat from 12 carbons to 18 carbons. Specifically we propose to investigate: 1) if making fat longer can alter the development of the brown fat organ; b) whether eliminating/increasing the capacity of elongating fat makes the animals fatter/leaner and c) less/more capable of making heat. To that end we will use cells and mice in which we will decrease or increase the capacity to make fat longer and then study the type of fat these animals make using very novel techniques that allow us to know all the types of fats that these animals will have. This information will inform us about which fats are good and which ones are bad and will allow us, for example, to modify the diets of humans and animals to ensure that they have the right type of fats to remain healthy.
Technical Summary
A fundamental question regarding the fate of cellular lipids, in particular free fatty acids, is how these lipids are targeted to various fates, either oxidation, storage, export, or incorporation into more complex membrane lipid species. In this proposal we aim to investigate if fatty acid chain length may affect the balance of oxidation vs storage in the highly metabolically active tissue, brown adipose tissue. We will primarily investigate the effect of lack of Elovl6, an enzyme responsible for elongating saturated and monounsaturated C12 and longer fatty acids up to C18. Thus the aim of this grant is to investigate the role of fatty acid chain length on control of fatty acid oxidation, focussing specifically on the fatty acid elongase Elovl6. Specifically we propose to investigate: 1) the role of Elovl6 in BAT differentiation, lipogenesis and function in vitro; b) the role of Elovl6 on energy balance and c) the effect of a lack of Elovl6 on maximal thermogenic capacity of BAT. To that end we will use both cellular and genetically modified animal models with a loss/gain of function in Elovl6 to investigate the effect of altered fatty acid chain length on fatty acid oxidation. In vivo models will be characterised using state of the art phenotyping including performing experiments at different temperatures in purposedly built facilities for thermogenic studies. Also we will use a molecular systems biology phenotyping approach using sophisticated lipidomic and transcriptomic profiling integrated into lipid metabolic networks using bioinformatic analysis.

| Description | The purpose of this study was to determine the role, if any, of fatty acid elongation in brown adipose tissue function. To that end we utilised the elovl6 KO mouse as a model with altered fatty acid elongation. Elovl6 was well suited to this roll, as it is expressed at much higher levels in brown adipose tissue than any other tissue. Elovl6 is regulated by thermogenic challenges and is upregulated by cold exposure and reduced by thermoneutral housing. Unlike previous studies of elongation and its roll in BAT function by studying Elovl3, Elovl6 do not have an obvious skin barrier defect, removing the confounding factor of poor thermal insulation on studies of BAT function. In line with the concept that Elovl6 was important for brown adipose tissue function we have identified that Elovl6 KO mice have a temperature dependent post natal survival. At 20oC, as reported previously, only 30% of Elovl6 KO pups survive to 3 weeks of age. However, this is not embryonic lethality as reported but instead is a temperature dependent post-natal lethality. The mendelian ratios of elovl6 KO mice can be corrected by breeding and housing pups at 24oC until weaning. This phenotype is reminiscent of other mouse models with defective BAT activity (UCP1 and PACAP). In adulthood, when housed at 24oC, mice lacking Elovl6 have a normal energy balance phenotype, with no differences in energy expenditure or food intake. However, Elovl6 was upregulated by cold exposure. In order to investigate this phenotype more fully we cold exposed Elovl6 KO mice. Mice lacking elovl6 exhibited both elevated energy expenditure and food intake (hypermetabolism). The mechanism behind this hypermetabolism remains to be elucidated but it is notable that the Elovl6 mice at both 24oC and 4oC exhibit increased expression of thermogenic markers in subcutaneous white adipose tissue (the location of so called beige adipocytes or 'Brite' cells) and to a lesser extent in canonical brown fat. Elovl6 KO mice have shorter chain fatty acids compared to WT mice. In WT mice, cold exposure leads to significant changes in fatty acid chain length in brown adipose, white adipose and liver tissue, but not in muscle. It is possible it is a direct regulatory event in which loss of Elovl6 results in an increased thermogenic response to cold via central mechanisms. However, a second possibility is that loss of Elovl6 has actually resulted in partially defective BAT. While this would seem paradoxical the UCP1 KO mouse exhibits elevated energy at sub thermoneutral temperatures, with the biggest discrepancy between WT and UCP1 KO mice being at 4oC - the lowest temperature studied. In mice, cold exposure has been demonstrated to dramatically increase levels of stearate in brown adipose tissue mitochondria, but not in muscle mitochondria, suggesting that altered mitochondrial function may contribute to the hypermetabolic phenotype. This aspect is currently under investigation. Finally, in addition to our specific investigations into the Elovl6 model we have carried out an extensive characterisation of adipose tissue lipid changes in various dietary and genetic models of obesity. So far, this has demonstrated that obesity and insulin resistance are associated with a lipid profile consistent with elevated SCD1 and Elovl6 activity, even after the composition of diets has been taken into account. This finding is consistent with the improved metabolic profiles of SCD1 (liver specific) and Elovl6 KO (whole organism) mouse models. |
| Exploitation Route | Our research has potentially important implications for the functional food industry. In particular margarines and yoghurt drinks have been an area of intense focus of research and marketing by many of the major players in the FMCG world (Unilever's Flora Pro Active and Raiso group's Benecol are the leading brands in the UK with combined sales exceeding 50 million GBP). While these products have focused on cholesterol lowering, there remains a large area to exploit in the area of functional foods with modified lipid compositions and improved metabolic outcomes. Making functional food claims is increasingly difficult following EU regulations, requiring greater scientific evidence. By providing a scientific basis to support the roles of specific lipids in animal and fly models will allow a more rational and targeted design for the development of new lipid-modifying functional foods. Whether Elongase 6 is a valid target for pharmacological agents is at present difficult to assess. Whole organism ablation of Elovl6 in mice improves insulin sensitivity, but while the mechanism remains unclear it is difficult to assess the likelihood of its efficacy and safety in humans. The research funded by this grant will provide substantial benefit to the research community working in both metabolic science and lipid biology. By identifying unique roles for the products of Elovl6 in control of energy balance and brown adipose tissue function will enable a better understanding of the impact of modified diets and a deeper understanding of what lipid changes mean. At present lipidomics is limited by a lack of functional information for many lipid species and on going work to improve this area of our understanding of metabolism is vital. The research related to the role of Elovl6 in regulating mitochondrial function will be invaluable to almost all areas of biology in which mitochondrial function is important. The alteration in fatty acid chain length in mouse models of obesity suggests a wider role of the elongases in the development of obesity related metabolic complications, this can potentially be targeted through dietary or pharmacological manipulation. |

| Results and Impact | EAS Advanced in Cardiometabolic Phenotyping in Rodents, Cambridge, UK Scientists from the University of Cambridge Biomedical Campus recently hosted the 2nd EAS Advanced Course in Cardiometabolic Phenotyping in Rodents. This 3-day course combines the state of the art facilities and expertise within the Disease Model Core at the WellcomeTrust-MRC Institute of Metabolic Science and the BHF Cardiovascular Centre. By integrating the expertise of these two specialized phenotyping centres, it provides a unique opportunity to learn about many of the skills required for in depth cardiometabolic phenotyping in small animals. The 32 participants, selected from centres across Europe, were treated to a programme of lectures, phenotyping demonstrations and data analysis sessions. A further 1-day workshop at the Sanger/EBI focused on mouse genetics, genetic engineering and informatics resources, highlighting the great opportunities provided by recent advances in genetic engineering purpose-made mouse models. Participants particularly appreciated the mixture of hands-on experience, combined with opportunities to ask detailed questions on how the techniques demonstrated should be used to suit their own research priorities. The course was sponsored by the European Atherosclerosis Society, British Heart Foundation and TSE systems, with organisation led by Prof Toni Vidal-Puig, Director of the MRC Metabolic Diseases Unit Disease Model Core. |

| Results and Impact | New protein could help burn fat faster By Meera Senthilingam, for CNN Updated 1109 GMT (1909 HKT) February 1, 2016 (CNN)-Research has shown that the fatter your body becomes, the harder it becomes to burn that excess fat. As people eat more fat than they burn, the body stores this excess in various parts of the body and as this storage increases, metabolism changes. However, all is not lost: a recent study by researchers at the University of Cambridge identified one of the proteins underlying this blockage of fat burn. "[This protein] is part of the system in place to help the body store more energy," says Antonio Vidal-Puig, Professor in Molecular Nutrition and Metabolism, who worked on the study. The culprit, called sLR11, was found to block the functioning of brown fat cells -- responsible for generating heat to keep us warm -- and instead help the body store fat more efficiently by preventing excessive heat generation. The benefits of brown fat There are two forms of fat cells -- brown cells and white cells -- and each plays a different role in our metabolism. While brown cells help generate heat, the white cells are responsible for the storage of fat -- or energy -- ready for release as needed. Levels of brown fat are known to be high in children but recent findings showing the presence of brown fat in adults, restored hope to use them as targets to treat obesity. According to Vidal-Puig, obese people have less brown fat than lean people -- and the same applies for diabetics. "If you can increase the brown fat in obese and diabetic people then you can improve their situation," says Vidal-Puig. This has been known for some time, but making it a reality has been a challenge. The newly discovered sLR11 protein could provide the answer. Removing the brakes Vidal-Puig compares the protein to the brake of a car, stopping the body from accelerating its production of heat. "Obese people have more of these brakes, preventing them from creating heat and therefore losing fat," says Vidal-Puig who has seen elevated levels of the protein in the blood of obese people included in his study. During the study, the team removed the gene producing this protein in mice and found they were able to burn calories faster. The researchers went on to examine the protein in humans and found that levels of the protein correlated with the total fat mass in a person. What's more, investigated levels in obese patients after they underwent bariatric surgery, and their weight loss was directly proportional to the decrease in levels of the sLR11 protein. "[Brown fat cells] are an attractive therapeutic target for weight loss, and for anti-obesity or anti-type 2 Diabetes (T2D) therapies," says Carole Sztalryd, Associate Professor of Medicine at the University of Maryland School of Medicine. Sztalryd's team also work on biological approaches to weight loss. More brown than white on the cards A common focus to tackle obesity has been on accelerating the production of brown fat cells so people burn more calories, but this new insight could achieve the same but through enhancing the activity -- and efficiency -- of cells. "We are stopping the brakes within the fat cells," says Vidal-Puig, as the mice in his study became protected from obesity with increased browning of their white fat cells. The team believe controlling energy storage will be a better approach to beat the obesity epidemic than controlling people's eating habits. But Sztalryd warns that additional studies are required to evaluate any clinical relevance,"New therapies are urgently needed for obesity. sLR11 seems promising but more work is needed to confirm if this is a relevant," she says. In the mean time, old tactics may be the answer and the more weight gained, the more exercise to be done. |

In spite of all the millions spent on diets, food and exercise, it may be that something as basic as temperature is the answer. We all have two types of fat, white and brown. White fat is inert and sits in deposits. Brown fat, which regulates babies' temperatures but is much harder to pinpoint in adults, can be activated to burn calories and generate heat. Studies show that exposure to low temperatures may encourage white fat to behave like brown fat. So while central heating ensures we move less and burn fewer calories, it's possible that lower temperatures might increase the activity of brown fat, which will burn our calories for us. |
