Abstract

Since it's first use nearly 400 years ago, microscopy has grown to be one of the most powerful and commonly used research tools by life scientists across the world. Using microscopes to image biological samples can provide a wealth of information ranging from understanding how tissues are organised during development and how this changes in diseases, to studying single proteins or DNA behave inside living cells. The types of microscopes that researchers use in the lab are often very advanced and complex in nature, and can require teams of experts to build and maintain them as well as to teach other scientists how to use them for their experiments. Moreover, analysing the data that research teams produce from these sorts of microscopes can be complicated. Nearly all Universities and research Institutes across the UK have microscopes in use within their research labs, but we currently lack good ways for these scientists from very different fields to share their imaging equipment, expertise and ideas. This sort of interaction is very important for knowledge exchange, training researchers and developing new ideas that will ultimately improve our research and mean the UK is at the forefront of this exciting technological area.

The purpose of this proposal is to support up a UK-wide network called BioImagingUK, which will provide a way for imaging scientists to interact, discuss newly emerging ideas and decide on key priorities that we need to work on to improve the quality of our research. A big focus of BioImagingUK will also be in supporting the technical and more junior imaging scientists and put in place important measures to help their career development and visibility across the UK research community. This is not only important for their careers, but also really helps the UK to retain exceptional talent to ensure we remain at the forefront of this internationally competitive scientific area. BioImagingUK will also help academic researchers to interact more closely with teams from associated Industries (such as companies who manufacture microscopes) to help build better relationships between the two sectors and identify new ways to develop novel technology together. Finally, this community network will also provide an important platform for bio-imaging scientists from across the UK to communicate their research to the public.

Technical Summary

Biological imaging is increasingly recognised as complex, intensive, and important for the achievement of national goals. The organisation and co-operation of the Bio-imaging community in other countries has been driven by, and continues to drive, a change in the landscape of infrastructure for biological imaging. The UK has also driven greater co-ordination of our research efforts through BioImagingUK, supported by our first network grant (Dec'14-Dec'17). BioimagingUK has thus far shown the potential power of co-ordinating and supporting the community and driving forward new ideas and initiatives that are important for this community and its success. To continue to ensure the success of BioImaging in the UK, we need to develop and support a more active network that builds on our success, and enables continued co-ordination of bioimaging over the next few years. The development of a national network to support Imaging Scientists is critical for the UK to remain at the forefront of biomedical research.

The purpose of this proposal is to renew support for the BioImaging UK network, not only to maintain our current efforts, but also to continue to drive our collaborative efforts in BioImaging across the UK to ensure the continued progress and achievement of biological and technical excellence in the UK. Through community meetings, workshops, surveys and the development of associated reports on these activities our goals are to 1) Develop a strategy for UK biological and biomedical imaging technology; 2) Develop meaningful career structures and evaluation criteria for BioImaging Research Technical Professionals (RTPs; Imaging Scientists). 3) Use BioImaging UK to develop and support training across practical aspects of imaging and image analysis and 4) Provide the community with up-to-date resources, organise and run community meetings and engage with global BioImaging activities for reporting back to UK community and stakeholders.

Planned Impact

BioImagingUK will provide a platform for scientists engaged in Bioimaging across the spectrum (from PhD students to research staff and Imaging Specialists) to access the relevant help, training and support. The Network will also provide a co-ordinated voice for the research and industry sectors to communicate knowledge and strategic ideas to the wider funding community. We will use this community-wide voice to raise the profile of early career researchers and provide them with opportunities to develop their personal networks and skills. Our plans for detailed reporting on better Career structure for Imaging Scientists in universities and research institutes will ensure continued support and career development for these essential, highly trained professionals.

A key goal of BioImagingUK is also to identify new strategic priorities, helping to drive investment in, and development of, imaging in the UK. Our provision of a single point of contact and information for the BioImaging community will enable this co-ordination of community ideas and enable better communication and input into UK-wide strategy for bioimaging in future. The 3 year proposal is built around specific timelines of engagement with different members of the BioImaging community with a view to regular reporting to stakeholders and constant feedback to the community via the website, Wiki and social media feeds. Such a co-ordinated activity is highly likely to drive our research efforts forward faster than through individual efforts, and more likely to lead to new ideas, sharing of best practice, new technologies and broader global recognition for the UK community in this important scientific area. BioImagingUK will be entirely distinct from, but highly complementary to, existing UK networks such as the TTLs, NIBBs and Physics of Life, which are focused on discipline-specific goals but encompass some aspects of Bioimaging. We will co-ordinate and engage with these existing networks to capture their involvement and needs in Bioimaging and ensure we are supporting emerging ideas from those networks as well as early career researchers starting out in Bioimaging fields. We anticipate significant synergy between existing networks and BioImagingUK that will ensure cross-fertilisation of ideas and consolidation of emerging needs within the Bioimaging community spanning a broad range of fields.

BioImagingUK also aims to actively engage with Industry throughout the course of the current proposal and beyond. These interactions will ensure we develop a complete picture of emerging bioimaging requirements within pharmaceutical and technology sectors and feed disseminate this to academic networks with a view to encouraging new collaborations and joint funding applications. We will also work with industry to determine their perception of the current skills gaps in Imaging-related areas and feed this into ongoing development of training workshops to enable academics to develop relevant skills for transitioning into industry. We will also seek to develop opportunities for researcher exchange and secondment between industry and academia to enable a better understanding of the challenges faced in each within the BioImaging domains and provide more direct mechanisms to interact and develop new mutually beneficial ways of working.
 
Title Helped to facilitate Overstory - a site-specific outdoor installation, giving a view into the microscopic world of trees 
Description Two leading contemporary artists, Ivan Morison and Heather Peak, designed two temporary artworks that were suspended above Broadmead in Bristol, 19-30 August 2022. OVERSTORY was a site-specific outdoor installation, giving a view into the microscopic world of trees and asking visitors to celebrate and consider how they are folded into our lives through their presence around us, their functions within our shared ecosystems, the stories they tell, and the role we can all play in creating and protecting the natural spaces in our cities. The artworks highlighted the importance of urban tree cover and encouraged the public to venture outside and discover their local parks, wildlife reserves, and older neighbourhoods to experience the diversity of native trees around Bristol and the stories they tell about the City. The artwork's microscopic imagery was created by working in concert with the Royal Botanic Gardens, Kew and Mesolab, at The University of Strathclyde's collections, captured using advanced technology; that reveals the complex organisation of tree life at the micron-scale. The BioImagingUK network facilitated this project by sourcing one of the high-quality tree sections using its extensive mailing list: The Lime tree specimen - this was captured using the Mesolens, a giant microscope designed for high-resolution imaging of large specimens. Its founder, Professor Gail McConnell from the University of Strathclyde and current Chair of the Royal Microscopical Society's Light Microscopy Section, explained "We are using the Mesolens to image many different kinds of biological specimens including plants, bacteria, and fungi; the bright features of the images are from naturally occurring molecules present in the tissues". Dr Liam Rooney, Fellow of the Royal Microscopical Society and Research Associate at the University of Strathclyde, added, "This Tilia specimen demonstrates the complex organisation of cells arranged into specialised layers of tissues. Its tiny holes are sections of long tubes that transport nutrients and water along the stem of the plant".

Description In response to Lockdown 1.0 in the UK, and because most researchers were unable to access the wet lab, we established and maintained a curated online resources page hosted on the RMS site. This included live streams, lectures, recorded talks and courses and was a great resource for many as there normal working lives were momentarily put on hold.

PI Contribution Georgina Fletcher, project officer, is one of five people on the RMS' Mentoring Working Group that started the Application Coaching and Personal Mentoring Scheme pilot. She has provided strong administrative support, sourced and adapted policies and forms from other Mentoring schemes, and used her extensive network to find suitable mentors for the program. We now have 8 pairs that have been matched and are in the process.
Collaborator Contribution The other four people in the Mentoring Working Group have helped shape, deliver and publicise this mentoring pilot.
Impact This is an active collaboration - we are currently designing feedback forms for all 8 pairs that will be used at 6 months and 12 months where we will capture any outputs or outcomes.

Description Formation of the UK Node of Euro-BioImaging (EuBI)

PI Contribution Following BioImagingUK-led community consultations, a community-written white paper and a review process in 2021 together with UKRI-BBSRC and UKRI-MRC, seven UK pilot sites were chosen and put forward to the EuBI call for Nodes in 2022. After a small revision, the UK Node of EuBI was accepted, ratified and began operation at the start of 2023. Georgina Fletcher is the UK Node Manager and coordinates user access and EuBI activities in the UK.
Collaborator Contribution EuBI evaluated and suggested minor revisions to the UK's application. They now provide the framework for external user application to our seven UK sites, including administrative support, communications, outreach, seminars, forums, meetings etc.

Results and Impact This meeting provided an opportunity for the UK Bioimaging community to discuss priorities and strategies in training, development, careers and ways to share knowledge across different disciplines. The session consisted of short talks from members of the BioImagingUK organising committee, BBSRC, and industrial/institute collaboration partners (Royce, RFI, Faraday) to update on progress, new opportunities and initiatives. There was an interactive Q+A sessions to encourage discussion and enabled emerging priorities and ideas to be highlighted. The meeting was open to everyone with an interest in bioimaging. Approximately 100-150 people attended.

Results and Impact Following on from the hugely successful RMS Covid safety LM, EM and Flow meetings in 2020, we have set up a virtual Coffee Club and invited all our members. Designed to be informal and a chance to interact with colleagues across the UK, this platform provides the opportunity to discuss a technical problem, a new technique, or something else entirely.
It's also providing a much needed support network for those people who manage core facilities on their own.

Results and Impact The hidden REF is a competition that recognises all research outputs and every role that makes research possible. Georgina Fletcher (BioImagingUK Project Officer) is a member of the committee and promotes the initiative amongst the UK bioimaging community. There were 120 submissions in 21 categories in the summer of 2021. Part of Georgina's role was to decide how to evaluate the entries. We used Significance, the scale and importance of the contribution; and Visibility, the degree to which the submission could not normally receive recognition from standard research-evaluation measures. She was also a Panel Advisor on the Practices panel which included Access to facilities, Standards and Community building. Like all evaluations, the Hidden REF framework will be improved over time as future iterations are run and the evaluation approaches, criteria and methods are adopted elsewhere. The Hidden REF will run again in 2023. There is interest in the framework from around the world, so it will run at different times in different countries.
Publications rarely name all of the people who make research possible, yet they were the basis for 97% of the outputs submitted to REF 2014. This is not the fault of the REF itself: the guidelines allow submission into a wide range of categories, from software to musical compositions. But with so much funding reliant on the results, universities are highly risk averse in what they will submit. Publications are well understood in academia, so they are almost the only output submitted to REF - and this means the work of many people goes unrecognised. The REF is invaluable to our understanding of research. The goal of the Hidden REF is to celebrate all research outputs and the people who make them possible.

Results and Impact Multimodal Imaging of Complex Organisms across Scales or MIcroCOSM is a suggested proposal for a UKRI-funded infrastructure. BioImagingUK, in partnership with UKRI, hosted an open online consultation/community workshop with the UK bioimaging community to gain feedback on plans for this infrastructure bid.
It was aimed at anyone interested in multiscale multimodal bioimaging spanning from in situ structural biology using cryo electron microscopy (EM), through to volume EM, light microscopy, X-ray microscopy and beyond, incorporating sample preparation, correlative workflows, big data handling, image analysis and AI approaches. That included but was not limited to: current and prospective users (discovery research and clinical research), technologists, industry (users and technology developers), data specialists (analysis, handling, archiving); software engineers; and funding bodies.
We had breakout groups on demand/access, technology, data as well as presentations to discuss the options and advantages/disadvantages of each model and the science it would enable.

Results and Impact The Royal Microscopical Society (RMS) holds Section Committee Meetings three times a year across the main branches of microscopy. Georgina Fletcher (Project Officer) has regularly attended the Light Microscopy, Electron Microscopy, Life Sciences, Early Career Committee and Atomic Force and Scanning Probe Microscopy Sections. In addition, three focussed interest groups (FIGs) on Image Analysis, Training and X-Ray Microscopy are also regularly attended. Updates of Network activities are provided by our Project Officer and discussion/input around topics are encouraged. We also use the power of our network to help members with their activities, for example we coordinated the distribution of an X-Ray Microscopy Usage Survey.

Results and Impact Our project officer, Georgina Fletcher gave a 10min presentation about the BioImagingUK network to the UK Light Microscopy Facilities Manager community. The purpose was to update the group about our current and future plans and to increase engagement. There was a significant increase in mailing list subscribers and twitter followers afterwards which we tentatively ascribe to the talk. Specific requests came in to use some of our material as well.

Results and Impact BioImaging UK convened a series of virtual meetings in April and June 2020 to build a community consensus for an Imaging Research Infrastructure (RI) in the UK. Several key questions were the focus of the discussions and the foundation for a community consensus:
A. Should there be a common, open-access Imaging RI in the UK? What form should this Imaging RI take?
B. How would an Imaging RI be financed? Would funding flow to individual Nodes of the infrastructure or exclusively to the central hub of the infrastructure?
C. How would access to the Imaging RI be controlled and managed?
D. How do we connect the facilities that have been built in the UK for managing and processing data to the Imaging RI?
