Abstract

Technical Summary
In Work Package (WP) 2 will provide mechanistic understanding of how genes regulate phenotypic traits and will thus enable the design of improved alleles to facilitate the control of important crop traits such as vernalisation and reproduction. A major challenge to predict and improve crop traits is understanding how gene output is regulated as plants adapt to changing external environments. In the previous GEN ISP we have discovered that chromatin-regulating factors and non-coding RNAs underlie many gene regulatory events during plant development and environmental responses. However the molecular mechanism by which these non-coding pathways regulate gene transcription and translation and how they are controlled in changing external environments are still largely unclear. Elucidating these mechanisms will have immense impact on improving crop yield by controlling vital processes such as flowering time fertility photosynthetic capacity and the circadian clock and to develop tools that will aid in transgene stability. Moreover the polyploid nature of many crops provides yield benefits over diploid relatives in terms of for example vigour and organ size and understanding the mechanisms by which individual homeologue expression is controlled will facilitate the ability to predict trait outcomes. In this work package we aim to dissect the mechanistic basis of chromatin-based gene regulation and its environmental modulation. The results directly link to WP 4 providing solutions to improve crop sustainability and resilience to abiotic stresses such as drought and heat. We will take a multiscale approach from studying single gene loci and gene clusters to the interactions within and between nuclear and plastid genomes and employ multidisciplinary approaches such as machine learning single-cell and single-molecule sequencing and cryogenic electron microscopy. Through these studies we will be able to precisely predict and edit gene output and crop traits.

Such single-molecule sensitivity combined with stepwise photobleaching was then be used to obtain the number of molecules in the FCA, VIN3 and VRN5 assemblies. |

The aim of the network is to support the circadian clock community inc reading bridges between chronobiological research and real-world impact. It aims to consolidate, engage and expand circadian biology research to establish and nurture robust cycles of discovery and impact. The team will also coordinate and lead training for the next generation of chronobiologists. BioClocks UK was started by volunteers from universities, the BBSRC-supported institutes JIC and IBERS, and the MRC Laboratory of Molecular Biology. BioClocks UK was recently awarded a 5-year BBSRC responsive mode grant to support its activities, held at the University of Leicester with grant PI Bambos Kyriacou. Antony Dodd leads and edits the newsletter for BioClocks UK. |
