A Relativistic Electron Diffraction and Imaging (RUEDI) Facility for Structural Dynamics on the Femtosecond Timescale

Abstract
Transformative innovations in the science and technology of personalised medicine, energy storage (grid and transportation), clean growth (energy generation, transformation and advanced manufacturing), and materials operating under extreme conditions will start from achieving atomic and molecular control of the fundamental (bio)-chemical interactions that determine each process. As Lord Kelvin famously said, "To measure is to know. If you cannot measure it, you cannot improve it". Here we introduce the vision of a national facility centred on the unique measurement capabilities offered by relativistic ultrafast electron diffraction and imaging (RUEDI). This globally unique facility would permit the direct observation of atomic/electronic motions directing the very chemistry that must be controlled to make the advances listed above. It would enable observations that are the essence of chemistry and by extension, the driving force for biological functions on their fundamental time and length scales.This would permit:
A determination of structure-function relations at the cellular level in biological systems as a means to better target drug development (such as for COVID-19) as well as create new biomimics, beyond mutagenesis, to harness biology in new ways.
Electrochemistry constitutes a significant fraction of the UK and global economy and RUEDI could be used to directly observe the molecular origins of the all-important electrical double layer in electrochemical systems and from it help develop new and improved energy storage systems.
Experiments to determine the fundamental mechanisms leading to functionality in catalysts, enabling a more rational hunt for new catalysts and providing the impetus for developing new photocatalysts that will drive future renewable energy sources.
The interplay between phonon and electron oscillations would also be directly observable and controllable, allowing RUEDI to uniquely probe new concepts in plasmonics and strongly coupled systems for electronic devices, sensors, probes and detectors that could transform the use of nanotechnology.
Experiments at the frontiers of spatial and temporal resolution using in-situ cells will permit a new fundamental understanding of how materials and devices perform under extremes of temperature, pressure, field and environment.
The fundamental structure-function relationships that the facility can uncover directly impact the EPSRC priorities of 21st Century Products, Digital Manufacturing, Sustainable Industries and New Industrial Systems. The RUEDI facility would also have direct impact on numerous research themes defined by EPSRC for growth, such as biophysics and soft matter physics, chemical biology and biological chemistry, energy storage, materials for energy applications, RF and microwave devices, and statistics and applied probability. The RUEDI imaging facility will be unique in the UK (and globally), and produce relativistic electron bunches at energies up to 5 MeV, at a repetition rate of 100 Hz. It is envisaged that each bunch contains up to 107 electrons, with the length of each bunch less than 100 fs. This provides enough electrons for diffraction patterns and images in a single-shot, with bunches short enough to resolve structural change on the timescale of biochemical processes. The key advantages in using relativistic electrons, as compared to the more common ~100-300 keV electron energies, are higher penetration depth and reduced space-charge effects, leading to shorter bunch lengths, greater transverse coherence, and more electrons per pulse, thereby enabling clear single-shot images to be observed. RUEDI therefore allows the evolution of structural changes in materials to be observed through time-resolved experiments. In this manner, this facility would be unique by allowing the all-important function of a material to be determined, rather than simply its static structure.

| Impact | The RUEDI Project operates as a unique opportunity to work with a number of stakeholders and partners with the potential to stretch into core areas of emphasis for BBSRC and MRC as well as providing overall development of new imaging Technologies for Biosciences and the key data for Systems Approaches to Biosciences. The synergy between these approaches for different research councils can also provide huge cross-cutting benefits for UK science and technology. The RUEDI facility will be organised to operate within the UK research community with the first five years spent on design, construction and testing the hardware to establish the basic framework for a user program. Key experiments identified at each of the scientific theme town hall events will involve stakeholders from the research community, from organisations noted including but not limited to the Faraday Institution, Crick Institute, UK Catalysis Hub, Henry Royce Institute and others. The facility will generate interest from international research stakeholders and partners. The initial plan for accommodating these users would depend on any agreements in place between their national research councils and UKRI. In future, transnational access arrangements could be evaluated for example, by joining European programme schemes, should the UK be eligible. A key part of the national facility will be support of UK industry in the use of these capabilities. While the exact details will be defined closer to when the facility is established, it is anticipated that support for industry stakeholders and potential partners is considered on a case by case basis with proprietary users operating within a full cost recovery model. As noted previously, an overarching Collaboration Agreement exists, with all new partners expected to formally sign an Accession Agreement. |

Professor Angus Kirkland will present 'Making Every Electron Count; Ptychography at Low Dose' The ISAMS-5 continues its tradition of promoting the scientific community engaged in cutting-edge research and development in scanning/transmission electron microscopy and electron energy-loss spectroscopy. Following ISAMS-5, UCI IMRI will host the UCI School of Transmission Electron Microscopy (UCI STEM) from December 6-8, 2023. This school will provide participants with a unique opportunity to learn from leading experts in the field and develop new skills in 4D STEM, spectroscopy, and in situ TEM. Participants will learn about the latest techniques and tools to advance their research and collaborate with other researchers in the field. The RUEDI facility will provide scientists in both academia and industry with unprecedented capabilities to investigate and understand molecular-scale dynamics in-vivo, in-situ and under operando conditions. Specifically, RUEDI will facilitate in-vivo and dynamical biosciences through: • In vitro and cryo imaging at whole cell scales - beyond medium energy tomography • Imaging of biological structures in a cellular context • Understanding infection and transport at high temporal and spatial resolution • Imaging nanoparticle drug delivery and release in cells In addition to the standard diffraction mode, the RUEDI facility will uniquely incorporate a set of electromagnetic lenses that will permit images to be acquired for the majority of in-situ, in vivo and operando experiments Working with an MeV beam, the thickness limitations that limit conventional TEM observations will be reduced, in particular in the design of thicker liquid flow cells that can operate with viscous liquids to mimic in vivo biological conditions. Here we describe the uniqueness of RUEDI and will collectively identify a representative set of challenges that illustrate the types of experiments that could be performed during the initial testing and operations phase of the RUEDI system. Professor Sven Schroeder, RUEDI Scientific Theme Lead, will host the meeting at the University of Leeds (Room 1.31, School of Mechanical Engineering). The RUEDI facility will open new perspectives for the control of chemical transformations for research across all science and engineering disciplines, including applications in energy, healthcare, environmental, materials, life sciences and medical research. The unique capabilities of the planned RUEDI facility will be described, with a focus on how RUEDI instruments will probe the chemical and structural dynamics on ultrafast timescales. These provide insight into the elementary steps of transformations on molecular time and length scales, providing a deep understanding of the molecular basis for chemical transformations, and by extension, biological function. FEMMS (Frontiers of Electron Microscopy and Materials Science) is a biennial meeting focused on the application of electron microscopy, primarily TEM, in materials science. The conference series primarily arose to satisfy the requirements of the electron microscopy community working on materials science problems, which grew explosively in the late 1980's and has expanded to researchers working with analytical periphery, novel instrumentation, tomography, operando and correlative microscopy, and advancing into the domain of ultrafast time-resolved microscopy. Traditionally, FEMMS brings together approx. 40 invited speakers plus one distinguished lecturer that are world-renowned experts, principal investigators and project leaders, to present their latest advances in the field of electron microscopy in the materials sciences. The scientific agenda of the meeting will continue to reflect the revolutionary advances in all aspects of high-resolution imaging with electron microscopy, including structural studies of dynamic multi-protein complexes using cryo-EM, analysis of sub-cellular architecture using cryo-electron tomography as well as novel methods for specimen preparation and correlative imaging. The meeting will also highlight accelerating advances in machine learning based methods for structure prediction and protein design. The developments at the interface between tools for high resolution imaging and machine learning methods have significant implications not just in structural and cell biology, but also in drug discovery. | Results and Impact | Dr Tim Noakes will be presenting a poster ''Photocathode and photoinjector options for the RUEDI (Relativistic Ultrafast Electron Diffraction and Imaging) facility' at this 3-day workshop next week. Development of photoinjectors is a critical research area for modern accelerators. Photoinjectors produce the high brightness electron beams that have enabled the realization among others of Ultrafast Electron Diffraction and Microscopy setups, X-ray Free Electron Lasers, Energy Recovery Linacs, high average current sources for bunched electron beam cooling and spin polarized electron beams used in colliders. All these accelerator machines rely on photocathodes to produce electron beams with precisely controlled temporal and spatial shapes, often with stringent requirements on emittance, temporal response, and spin polarization. This 3-day workshop hosted by Brookhaven National Laboratory (October 3-5, 2023) will explore the current state of the art in accelerator photocathodes, from operational, theoretical and a materials science perspective, aiming also at identifying directions for future research and new opportunities for collaboration. | Results and Impact | Dr Julian McKenzie will be presenting 'Establishing a Relativistic Ultrafast Electron Diffraction and Imaging (RUEDI) Facility in the UK'. https://conferences.pa.ucla.edu/PAHBB23/index.html The ICFA-supported workshop series on the Physics and Applications of High Brightness Beams is traditionally a meeting ground for discussion of high brightness beam production, manipulation, and acceleration in state-of-the-art systems ranging from cutting-edge RF accelerators, to very high-field plasma-based schemes. The resultant beams provide the underpinnings of new scientific instruments such as X-ray free-electron lasers (XFELs) and TeV-class linear colliders. The workshops also provide an essential opportunity to guide the full ecosystem of beam physicists along with the advanced user communities who utilize the newly emergent instruments arising from high brightness beam research. In the upcoming version of this workshop series, we will concentrate on a particularly compelling example of the burgeoning field of new instruments based on advanced, high-field accelerators and high brightness beams. This is the 5th generation light source, which seeks to merge two compelling new instruments - next-step, compact XFELs, and advanced, high-field accelerators. This merger has produced a hybrid field, which has by now many promising incarnations. The present workshop will concentrate on the physics of such systems, which are now proceeding towards realization using approaches ranging from cryogenic RF to wakefields in plasmas and structures. These initiatives are strongly motivated by synergistic application to the energy frontier, as 5th generation light sources may also be fruitfully seen as stepping stone projects which demonstrate beam physics feasibility issues needed to proceed to future linear colliders based on advanced concepts. Such ambitions require a continuous re-examination of progress in the fields of high brightness beam physics, advanced acceleration, and their nexus in applications. This is precisely the mission accepted at this workshop, and we look forward to seeing you in this year's meeting, taking place at the Kursaal venue in San Sebastian, Spain, June 19-23, 2023. This edition of the workshop will emphasize two emerging trends: first, the vital involvement in the development of the field through student and post-doctoral training through a dedicated half-day session sponsored by the NSF STC Center for Bright Beams); and the key involvement in the future user communities of 5th generation light sources. | Results and Impact | The Relativistic Ultrafast Electron Diffraction & Imaging (RUEDI) Facility project invites you join us for a Town Hall Event on the RUEDI Quantum Materials Scientific Theme on Wednesday, September 28th 2022. Dr Will Bryan, RUEDI Scientific Theme Lead, will host the meeting at the Harwell Campus in Oxford. The RUEDI facility will open new perspectives for research across all science and engineering disciplines, including applications in energy, healthcare, environmental, materials, life sciences and medical research. An emerging field at the intersection of condensed matter physics, quantum optics and laser physics, the umbrella term "Quantum Materials" unifies a diverse range of concepts across science and engineering. Such processes are typically strongly correlated, far from equilibrium and non-thermal in nature. From process involving charge, electric and magnetic fields, via quasi-particle dynamics and electron correlation dominated processes, to cavity QED, Quantum Materials has a broad scope, and can be considered highly relevant to societally important considerations such as energy generation, storage and transportation, quantum communication, and very low energy electronics. |

| Results and Impact | The presence of RUEDI at Daresbury Laboratory will present opportunities to utilise the facility for activities to engage the general public with both the specific science performed at RUEDI and with accelerator science in general. High-quality public engagement is already regularly performed at Daresbury through collaborations between the Accelerator Science & Technology Centre, the Cockcroft Institute and the STFC professional on-site public engagement team. The unique nature of RUEDI within the UK will provide strong enhancement to the existing programme and will make RUEDI a valuable tool to capture the public interest in science and inspire the next generations of young scientists. STFC operates a public engagement programme known as the "Wonder Initiative" [1] specifically targeting public outreach in the most socially deprived areas of the UK, especially 8-14 year olds and their families. People and schools in these areas typically have less access to Science, Technology, Engineering and Mathematics (STEM) engagement opportunities, which has been directly correlated with lower uptakes of career opportunities in STEM amongst the populations of these areas. The wonder initiative is based on the ASPIRES research project [2] and the concept of "Science Capital", described as the extent to which people are exposed to scientific knowledge, values, experiences and attitudes in daily life. Daresbury is close to several areas that fall inside the Wonder Initiative's purview. The addition of RUEDI to the site and its use in outreach events and activities will provide an opportunity to raise the science capital of the entire northwest region. Public opportunities to engage with RUEDI will occur at scheduled events of two types; those aimed at public sentiment and those aimed specifically at education. Public sentiment is engaged at Daresbury through activities including the regular "Talking Science" lecture series [3], visits to external events such as festivals where volunteers speak to the public about the science at the lab, and laboratory open weeks on a 4 yearly cycle where the laboratory site is opened up to several thousand of the general public on a weekend and they are invited to tour the facilities including accelerators on site - where safe to do so. It is expected that the science performed using RUEDI would make strong material for public lectures and that the instrument itself would be open for public tours during open weeks. RUEDI's involvement in educational outreach will be regular in schedule and varied in nature. Daresbury staff regularly visit schools and the laboratory site regularly hosts visits from schools, the largest of which is the yearly Cockcroft institute Accelerator and Particle Physics Master Class (APPMC) [4] which has historically engaged approximately 250 students per event. Many of these visits, and especially the APPMC, have involved accelerator facilities being opened for tours as well as talks on the science performed within and hands-on activities explaining how the facilities work. RUEDI and the science performed at the facility will feature heavily in future iterations of these visits. |

| Results and Impact | The TEM-UCA Summer Workshop Transmission Electron Microscopy of Nanomaterials is devoted to review the advancements of Scanning/ Transmission Electron Microscopy, both imaging and spectroscopic techniques, in the characterization of a wide range of nanomaterials. The Workshop is limited to a maximum number of 24 attendees. Our attendees are people currently working or interested in the field of nanomaterials and their characterization by TEM Professor Nigel Browning will be presenting at the above event. Speakers will illustrate how TEM techniques as High Resolution Transmission Electron Microscopy, High Angle Annular Dark Field imaging, Electron Tomography, Electron Energy Loss Spectroscopy, and Cs corrected S/TEM studies can be combined to obtain structural and chemical information of the analysed materials at the atomic scale. Some examples illustrate the practical aspects of nanomaterials characterization and its applications in different fields. The participants will be divided in 2 groups to participate at computer labs in which they will get trained in the practical use of different software applications in the field of TEM. The topics covered in these computer labs are: High Resolution Electron Microscopy (HREM), Image Processing and Image Simulations. Recent advances pioneered by LANL's Accelerator Operations and Technology (AOT) division enable production of ultrashort, ultrabright, high-energy electron bunches amenable for use in a relativistic electron diffraction (MeV-UED) instrument. This workshop endeavours to provide an opportunity for both the material science and accelerator physics communities at LANL and beyond to come together to identify key questions in structural dynamics that can be addressed with this emerging technology. Emphasis will be placed on identifying current state-of-the-art technological advances and exploring future opportunities for next generation MeV-UED machines.
