Physical Organic Chemistry: Opportunities in Synthesis, Materials and Pharmaceuticals

Abstract

This application aims to catalyse and sustain a new dimension in UK research capability in physical organic chemistry. Our strategic alliance in physical organic chemistry will provide a unique continuum of expertise to tackle research opportunities in areas as diverse as materials chemistry, synthesis methodologies and pharmaceutical discovery and development. It will have the capability to address issues from solid-state to solution and gas-phase, from small molecules to biopolymers, and from nanoscale to pilot plant. We focus on topics of international significance to industry worldwide as well as to academic chemistry, that will help to (i) drive the creation of 21st-century electronic materials, devices and technologies (ii) understand and exploit methodologies for assisting chemical reactions with the potential to revolutionise energy use in chemicals and pharmaceuticals industries (iii) provide new and more effective medicines through understanding molecular recognition in pharmaceutical systems including drug-receptor, drug-drug and drug-carrier complexes. Its importance is underlined by the initial substantial support from diverse sectors of the chemicals and pharmaceuticals industry that we have so far put in place. It will initially lead to 26 new appointments, and we look forward to even more dynamic growth as the program unfolds.

Description This grant has provided a framework for development of many facets of physical organic chemistry, from advanced materials to reactions mechanisms to breakthroughs in crystallisation and manufacture.
The award (EP/E036244/1, Physical Organic Chemistry: Opportunities in Synthesis, Materials and Pharmaceuticals) has contributed to a number of substantial follow on activities. The strong focus on crystallisation and investments in single crystal diffraction contributed to the strengths that underpinned the development of CMAC as a collaborative manufacturing research centre for continuous crystallisation and processing of pharmaceuticals, co-created with GSK, AZ and Novartis. Glasgow University have been collaborators on the subsequent critical mass awards that include: EP/I033459/1, EPSRC Centre for Innovative Manufacturing for Continuous Manufacturing and Crystallisation; EP/K503289/1, Doctoral Training Centre in Continuous Manufacturing and Crystallisation and EP/P006965/1, Future Continuous Manufacturing and Advanced Crystallisation Research Hub. Moreover, the early career researchers appointed into SIPBS have been able to utilise the equipment base to accelerate their career, for example, Dr Iain Oswald secured an EPSRC early career manufacturing fellowship EP/N015401/1, Pressure-Induced Nucleation for the Continuous Manufacture of supramolecular assemblies. In addition we have secured additional investments to augment our capabilities in surface analysis of pharmaceutical systems via EPSRC and Wolfson Foundation awards for fast-scan AFM and ToF-SIMS.
It also led to the Prosperity Partnership with GSK and the University of Nottingham that starts in 2019.

It provided training for many early career researchers and particularly to the appointment of at least one additional academic, Dr. David Nelson, recently promoted to Senior Lecturer, and more than 26 PhD students at the University of Strathclyde, notably co-funded by GSK and the University of Strathclyde, engaged in physical organic chemistry. A key example of international success in this follow-up is Joshua Barham, who won the world championship of chemistry PhDs, namely the Reaxys Prize, in 2017 for his studies on site-selective late-stage functionalisation of complex moleculs of biological interest. Joshua is the first such winner to be associated with a Scottish University.[https://www.elsevier.com/solutions/reaxys/reaxys-phd-prize]. It is clear that this award has been the springboard for a number of important interactions between UK chemistry and pharmacy and the pharmaceutical industry. Through it, we compete at a higher international level.
Exploitation Route The findings are being taken forward in collaborations with pharmaceutical companies for clean manufacturing processes.

Description The summary below includes impacts on academic and non-academic spheres. Our Physical Organic Chemistry (POC) grant had three streams and so the following commentary follows similar lines. Impacts have been numerous, building on the POC award, extending to wider applications of diffraction to support structural investigations of organic pharmaceutical solids. These include investigations of novel molecular solids with international groups (Nature Materials volume 20, pages403-409 (2021); Mechanistic insights of evaporation-induced actuation in supramolecular crystals | Nature Materials). International impact is seen via Dr Andrew Urquhart who was employed under the award has taken up an academic post in Denmark and is Associate Professor in the Dept of Health Technology at the Technical University of Denmark (DTU). However, other technical areas of interest established under the POC on surface analysis have continued to thrive yielding high impact multidisciplinary scientific papers (e.g. Nature Chemistry volume 12, pages 914-920 (2020); Olanzapine crystal symmetry originates in preformed centrosymmetric solute dimers | Nature Chemistry). We have continued to successfully build our pharmaceutical materials theme with strategic equipment funding from EPSRC looking at compaction simulation for crystalline and amorphous systems (EP/S02168X/1; 2019-22; PI: Florence, Strathclyde) and novel in line flow measurements of particle formation processes (EP/T006331/1; 2020-2023, PI Prof Meldrum, Leeds University). Also new international collaborations have been developed expemplified with the recent EPSRC International Centre to Centre award to CMAC at Strathclyde for the Digital Design and Manufacture of Amorphous Pharmaceuticals with University of Copenhagen in Denmark and TU Ghent in Belgium (EP/W003295/1; 2022-25; PI Florence). These projects have attracted industry partners including GSK, AstraZeneca (AZ), Lek Pharmaceuticals and CCDC. Our work on scattering studies of molecular solid structures from the POC has extended to amorphous systems and recent award of a KTP with AZ to implement PDF measurement in their internal development pipelines highlights the industrial relevance and ongoing translation of our early research into industrial practice is actively continuing and will lead to further impact in the coming years. The NMR and microwave equipment that was contributed through this award allowed physical organic chemistry to flourish and the equipment helped to train many highly successful PhD students. One example is Dr. Franziska Schoenebeck, now Professor at RWTH University, who is Associate Editor of Journal of the American Chemical Society and a leading international light in experimental and computational mechanistic chemistry. Another example is David Nelson, who completed his PhD here in POC and in 2014 was appointed as a Chancellors Fellow in Strathclyde. He has since been promoted to Senior Lecturer and is a highly active member of the POC community internationally, both for his own research and for his international collaborations, notably in Spain. The grant also led to international collaborations, e.g. between Prof Murphy and Prof Chiba (NTU, Singapore) which was supported by funding from The Royal Society and oversaw exchanges of PhD students between Singapore and Strathclyde. The POC grant also has led to ongoing international recognition of our role in POC. Thus, this year, 2022, Prof Murphy will be a Plenary Speaker at the Reaction Mechanisms Conference, Boulder, Colorado, and Keynote Speaker at the 25th International Conference on Physical Organic Chemistry (ICPOC-25) in Hiroshima, Japan as well as Invited Speaker at the Pacific Symposium on Radical Chrmistry (PSRC-12, Kyoto, Japan) (now postponed until 2023). High impact papers on POC and mechanistic themes are developing high citation numbers e.g. J. Am. Chem. Soc., 2016, 138, 7402-7410. In industry, our successes in POC were influential in the development of (i) the ongoing GSK-SU joint PhD programme (led in SU by Prof Kerr) which has been outstandingly successful in providing very large numbers of PhD students for UK plc and also (ii) the ongoing EPSRC co-funded Prosperity Partnership between GSK, University of Strathclyde and University of Nottigham EP/S035990/1 'Accelerated Discovery and Development of New Medicines: Prosperity Partnership for a Healthier Nation' The Glasgow University component of the POC award led to the appointment of Dr Goetz Bucher (https://www.gla.ac.uk/schools/chemistry/staff/goetzbucher/) and the purchase of equipment (e.g EPR and high end UV-vis-NIR and fluorescence spectrometers). Following the completion of the grant, Physical Organic Chemistry remains a strong and flourishing sub-discipline within the School as indicated by our more recent appointments (Professor Peter Skabara, Professor Dave Adams). Researchers in post prior to the award (Professor Graeme Cooke, Professor Richard Hartley) have benefited significantly from the equipment associated with this grant. Furthermore, the excellent equipment base the grant provided has allowed the School to appointment of Dr Stephen Sproules (EPR specialist) and Dr William Peveler (fluorescence sensing).
