Advanced laser-ion acceleration strategies towards next generation healthcare

Abstract

The project aims to reach an important milestone towards the development of innovative healthcare technologies: all-optical delivery of dense, high-repetition ion beams at energies above the threshold for deep-seated tumour treatment and diagnosis (~200 MeV/nucleon). Driven from an immediate impact in accelerator science, the flexibility and compactness of the planned solutions, jointly with other potential advantages of laser-based systems, could revolutionise cancer treatment methods.

The extreme conditions reached during the interaction of an ultra-intense laser pulse with matter can lead, if suitably controlled, to the rapid acceleration of beams of ions with unique properties. The study of these laser-initiated acceleration mechanisms, and the characterization and optimization of the ion beams produced, have been, over the past decade, one of the most active and fruitful areas of high-field science. UK scientists have been at the forefront of the development of laser-driven ion sources. During the final stages of LIBRA, novel acceleration mechanisms have emerged, mostly based on the enormous pressure exerted by powerful laser pulses onto irradiated matter, which promise a step change in particle acceleration capabilities.

A key area of application of high-current ion beams (proton and carbon) is in cancer therapy. Through a series of coordinated and interlinked activities over a 6 year period, we aim to advance laser-ion acceleration to the point at which laser-driven beams will become a serious alternative to conventional RF accelerators for medical therapy. Besides offering a reduction in cost and footprint of particle therapy centres (major factors limiting their growth worldwide), a laser-driven approach would offer a number of advantageous features currently unavailable: on-demand switching between species (H and C, with options for other light ions such Be and Li) and energy control; enhanced diagnosis, with synchronized proton and x-ray pulses, and on-site isotope production for Positron Emission Tomography. Our ambition is for UK science to play a leading role in the development of high-energy ion sources, by capitalizing on the exceptional pool of expertise available to our consortium.

Planned Impact

The proposed programme has wide-ranging potential academic, economic and societal impacts. The programme addresses a key societal grand challenge, to advance the development of innovative healthcare technologies. Through the development of laser-driven ion accelerators to achieve this, major applicative breakthroughs in other sectors, including industry, energy and security, can be expected, due to the unique and transformative potential of ultrashort pulse laser-driven sources.
The short term immediate impacts are in the area of hadrontherapy / oncology and imaging. This has a high impact on society and quality of life. Protontherapy has in recent years become a key part of the national strategy for improving cancer treatment in the UK, with £150M Government investment in two new centres. Compact laser-based ion accelerators of the type that we are developing in this project could have major impact as a next generation, cost effective technology for use in cancer therapy. The same laser drivers have the added value of enabling on-site production of positron emission tomography (PET) isotopes and other radioactive tracers for use in medical diagnostics, and by changing the interaction conditions, can be used to drive high energy electron and photon beams for medical imaging. Furthermore, the sources can be used to produce multiple types of radiation in synchronised pulses, which would enable multiple types of imaging and therapy at the same time. This could have a transformative impact on cancer detection and treatment in the future.

The unique properties of the high energy laser-driven ion beams that we are developing are therefore likely to have widespread application in other fields. Conventional accelerators are widely used in many industries (for example in the development of electronics and in lithography and implantation applications), in energy (acceleration-driven spallation reactors) and in security (for the detection of illicit materials). Compact laser-based sources are likely to find niche applications in these areas, and may, in the longer term, become a displacement technology. As such the work undertaken in this programme will deliver benefits to related industry and ultimately the UK economy.

The proposed research is at the forefront of high power laser science and is one of the core topics driving the development of new international-scale high power laser facilities (such as the Extreme Light Infrastructure project in Europe). It will provide a pool of trained scientists, some of which will go on to take on leading roles in the many international laser projects being developed, which others will take up positions in industry. The research enabled by the proposed programme will therefore provide lasting positive benefits to society and to the economy.

Description In the course of the project we have made significant progress in key areas, toawrds the delivery of ion beams with characteristics suitable for use in Cancer therapy.
1) We have advanced a number of ion acceleration mechanisms based on the interaction of high power lasers with appropriate target media. Among these, we have demonstrated that, by controlling some key characteristics of the laser pulse, it is possible to accelerate Carbon ions to significant energies through the strong pressure applied by an intense laser pulse onto an ultrathin Carbon foil, with a key paper published in 2017 in Physical Review Letters. Furthermore, we have developed a technique, which, by exploiting large, highly transient electric fields generated by intense laser interaction, it is possible to improve the properties of laser driven ions, leading to highly collimated beams with small energy spread, highly suitable for being transported to an application site and for developing compact beamlines for medical applications.
2) We have progressed significantly our understanding of how an ultraintense laser pulse interacts with dense matter. In particular, we have elucidated the interaction of a laser pulse with a foil heated by the laser and becoming transparent to light due to its expansion. Recent publications in Nature Physics and Nature Communications showed that the laser pulse can drill a hole through the foil and that diffraction through this self-induced aperture controls the movement and structure of the particles being accelerated by the laser. This regime of interaction has led to very significant advances in the energy of the protons that are accelerated, which has recently reached 100 MeV - a very significant threshold in view of cancer therapy application, as these energies can already penetrate several centimeters within the body.
3) We have demonstrated that meaningful experiments investigating how the laser-driven ions interact with cellular media can be carried out in a laser-plasma environment. We have collected a significant amount of cell biology data, which is being compared and assessed against reference data obtained with conventional, longer pulse accelerators.
Exploitation Route The main motivation for this work is to enable the uptake of laser based technology in medical therapy and to provide new capabilities for radiobiological studies.
Our results in ion acceleration have received very significant exposure through publication on high-profile journal (Nature group journals and Physical Review Letters) and prestigious conference presentations, and are already influencing research in the field internationally, e.g. through the inclusion in future plans for major international developments (ELI Beamlines, ELI-NP). While analysis and publication of our radiobiology results is still ongoing, we are confident that the extensive body of research we have carried out in this area will have significant impact in determining future applicative uptake of laser-driven ion beams.

Description The project has developed novel concepts for ion acceleration which have had a very significant impact within the field of innovative accelerators. As an example, the achievement of near 100 MeV proton energies through an hybrid acceleration method, as reported in the very highly cited paper by Higginson, Nature Comm., 9, 724 (2018), has established itself as a milestone in the development of laser-driven proton accelerators, and has attracted significant interest across other research communities, in view of the potential medical interest of protons in this energy range. Similarly, work on carbon acceleration via radiation pressure acceleration (McIlvenny et al, PRL, 127, 194801, 2022) has received significant attention, being featured in several broad-audience physics magazines and newsletters. Our research and results have also impacted on the development of new projects and facilities. Through our experimental research , we have contributed to the scientific and applicative case for the development of STFC's Extreme Photonics Application Centre, a new national facility aimed to develop and apply novel accelerators and particle sources to support UK science, technology, innovation and industry, as well for the VULCAN 20-20 project, a major upgrade of the existing VULCAN system at STFC's Central Laser Facility which will provide world-leading scientific capabilities in the UK . Internationally, techniques developed within our project for all-optical proton beam collimation (described in Kar et al, Nature Comm, 7, 10792, 2016) have been integrated in the activities of the pan-European Extreme Light Infrastructure facilities, including on the ELIMAIA proton beamline, the first open user irradiation beamline employing laser-accelerated particles, where our technique will be key to increasing the proton flux and dose deliverable to users. Our project has also explored extensively the application of laser-driven proton beams to cell radiobiology, demonstrating the feasibility of laser-based, biologically meaningful experiments of potential relevance to future forms of therapy. This has contributed to raising interest in the future medical application of laser-driven beams, particularly at the extremely high dose rates we have now achieved, and to providing a motivation for the development of a dedicated UK facility for radiobiological studies. STFC has recently funded a feasibility study for the development of a Ion Therapy Research Facility, with a laser-based approach as one of the potential technologies explored, through the LHARA (Laser Hybrid Accelerator for Radiobiological Applications) project of which members of our consortium are part. Also in this area, our work is having influence on the activities of the ELI facilities, where flagship experiments in radiobiology on ELIMAIA are being planned based on our research findings. The long term aim of these projects and infrastructures is to achieve impact of high societal importance by developing and promoting advanced, more efficient forms of cancer radiotherapy. Finally, another important impact of our project has been in training a cohort of doctoral and postdoctoral research scientists which are now providing high quality contributions to major national and international research laboratory, academia and industry.

Title BEAM FOCUSING AND ACCELERATING SYSTEM 
Description A system for focusing and accelerating a beam of electrically charged particles, for example protons. The system comprises: a beam generator; a charge pulse generator;and a focusing and accelerating device comprising a body with a core. The body defines a charge path extending along the body and beam generator directs a beam of electrically charged particles through the core. The charge pulse generator simultaneously delivers charge pulses to the charge path. The charge path may be helical in shape. Movement of the charge pulse along the path creates an electric field that simultaneously accelerates and focuses the beam.
