Photo-Electro: Transforming Chemical Synthesis, Discovery and Manufacture

Abstract

Our vision is to use continuous photochemistry and electrochemistry to transform how fine chemicals, agrochemicals and pharmaceuticals are manufactured in the UK. We aim to minimize the amount of chemicals, solvents and processing steps needed to construct complex molecules. We will achieve this by exploiting light and/or electricity to promote more specific chemical transformations and cleaner processes. By linking continuous photochemistry and electro-chemistry with thermal flow chemistry and environmentally acceptable solvents, we will create a toolkit with the power to transform all aspects of chemical synthesis from initial discovery through to chemical manufacturing of high-value molecules.

The objective is to increase efficiency in terms of both atoms and energy, resulting in lower cost, low waste, low solvent footprints and shorter manufacturing routes. Historically photo- and electro-chemistry have been under-utilised in academia and industry because they are perceived to be complicated to use, difficult to scale up and engineer into viable processes despite their obvious environmental, energy and cost benefits. We will combine the strategies and the skills needed to overcome these barriers and will open up new areas of science, and deliver a step-change (i) providing routes to novel molecular architectures, hard to reach or even inaccessible by conventional methodologies, (ii) eliminating many toxic reagents by rendering them unnecessary, (iii) minimizing solvent usage, (iv) promoting new methodologies for synthetic route planning. Our proposal is supported by 21 industrial partners covering a broad range of sectors of the chemistry-using industries who are offering £1.23M in-kind support.

Therefore, we will study a broad range of reactions to provide a clear understanding of the most effective areas for applying our techniques; we will evaluate strategies for altering the underlying photophysics and kinetics so as to accelerate the efficiency of promising reactions; we will transform our current designs of photochemical and electrochemical reactors, with a combination of engineering, modelling and new fabrication techniques to maximize their efficiency and to provide clear opportunities for scale-up; we will exploit on-line analytics to accelerate the optimisation of continuous photochemical and electrochemical reactions; we will design and build a new generation of reactors for new applications; we will identify the most effective strategies for linking our reactors into integrated multi-step continuous processes with minimized waste; we will demonstrate this integration on at least one synthesis of a representative pharmaceutical target molecule on a larger scale; we will apply a robust series of sustainability metrics to benchmark our approaches against current manufacturing.

Planned Impact

Our Programme, Photo-Electro, will have substantial impact on chemists in academia and industry because we will demonstrate the value of continuous photo-, electro- and thermochemistry to make complex molecules. Making such molecules is a key activity in a wide range of UK chemistry-using industries. These include pharmaceutical and agrochemical manufacture as well as companies making speciality chemicals for fragrances and many other end products. Such industries are under constant regulatory and financial pressure to develop processes that are environmentally and economically more efficient than current methodologies. Therefore, our approach of replacing toxic and/or wasteful stoichiometric reagents by light, electricity and heat, has great attractions as demonstrated by the letters of support from our 21 industrial partners.

What makes our programme different is that we are using light, electricity and heat together to deliver integrated processes to carry out multi-step assembly of complex molecules. At the same time, we are designing and building new continuous reactors which, by the end of the Programme, will be available to non-specialist chemists in industry and academia. The designs will be informed by computer fluid modelling and chemical engineering so that we maximise the efficiencies of both mass and heat transfer. Our reactors are intended to be both reliable and scalable so as to simplify their eventual introduction into commercial chemical production. We have a robust strategy for impact. Our industrial partners will form a stakeholders' forum to engage with our research right from the start. The Forum is intended to be inclusive and new members will be warmly welcomed at any stage of our programme. Photo-Electro is particularly timely as it will deliver new methodologies which can increase the competivity of UK chemical manufacturing. And, in this period of post-referendum uncertainty, increasing that competivity must be a national priority.

Results and Impact Prof Sir Martyn Poliakoff has lectured "Towards Greener Chemistry" and "Continuous Green Chemistry" on local, national, and international levels. Since the start of PhotoElectro, Prof Poliakoff has given over 30 outreach lectures at venues including but not limited to: The Stokes Society, Imperial College, ENEQUI XXI (San Paulo, Brazil), industries including Givauden, Novartis and EPFL (Switzerland), and the Ashbourne Festival to name a few. Additionally, he was invited to give the James Walker Memorial Lecture and the University of Edinburgh. Each lecture was aimed at disseminating the importance and progress within green chemistry. Interest within the subject was increased after the lectures.
In 2020 Sir Martyn gave an on-line talk "The elements of Chemistry" to ca 200 sixth-formers via the West Midlands Chemistry teachers' Forum in Birmingham in November. He also gave a video lecture "Frontiers of Green & Sustainable Chemistry" via University of Nottingham Nignbo Campus in November. This lecture increased interest in photo-chemistry and exposed undergraduate and post-graduate researchers to the field. He also presented "Photochemistry, Spectroscopy, and Supercritical Fluids: From organometallic Alkane Complexes and C-H Activation to the synthesis of anti-malarial drugs" at ESPCI, Paris and the University of Sheffield. "Approaches to Scaling-up linked Continuous Flow Photo-,Electro-and Thermal-chemistry at Scale-New Reactors and New Approaches-From Picoseconds to Tonnes" Virtual Sustainability Workshop, University Jaume, Castellon, Spain.

Results and Impact Prof Richard Brown presented "Preparative Electrosynthesis Using Undivided, Extended Channel Length, Flow Reactors", "Practical Organic Electrosynthesis in Laboratory Flow Electrolysis Cells with Extended Channels: Milligrammes/Hour" and "Approaches to Preparative Electrosynthesis" at conferences and seminars across the UK. The lectures were aimed at increasing knowledge of and interest in electrochemistry. Additionally in 2018, that lectures were given to GSK (Stevenage, UK), Charles River (Chesterford Research Park, UK), and the University of Greenwich. In 2021 Prof Richard Brown presented 'Organic Electrosynthesis in Flow Reactors with Extended Channels' to the iPRD (Institute of Process Research and Development) in April at the Industrial Club at the University of Leeds via Microsoft Teams. In Oct 21 Richard Brown spoke in the Flow Chemistry session at the Lorentz Center Workshop on Electrifying Chemistry: from fundamentals to industrial application held in Leiden, the Netherlands.

The team presented an exhibit entitled 'Green Light for Chemistry' which highlighted the need for change within chemical manufacturing and how PhotoElectro's research is trying to make chemical manufacture more sustainable through the use of reactors designed to use photo- and electro- flow chemistry.
