Technical Summary

The Confidence in Concept scheme is a key part of MRC’s translational research strategy and provides annual awards to institutions, to be used flexibly to support the earliest stages of multiple translational research projects. The award can be used by the institution to support a number of preliminary-stage translational projects. The projects supported should aim to provide sufficient preliminary data to establish the viability of an approach –– before seeking more substantive funding. It is intended to accelerate the transition from discovery research to translational development projects by supporting preliminary work or feasibility studies to establish the viability of an approach.

Description A total of 56 derivatives from Series A and Series B have been synthesised, leading to an improvement in the REF for labelling Rab27a up to 140x compared to glutathione. These molecules are the starting points for on-going ligand growth/elaboration work either through secondary screening or PROTAC development.

Title ICiC 6 - Tate - Methods for assessing reaction between covalent fragments and target molecules. 
Description New methods have been developed to assess the rate of covalent modification of a protein using a fluorescence anisotropy readout (rather than fluorescence intensity in the original QIT assay). This new method allows from more sensitive detection of labelling in more dilute protein samples. Secondary confirmation assays (intact protein mass spectrometry, peptide mass fingerprinting, differential scanning fluorimetry) have also been integrated into the QIT screen. These methods have allowed assessment of newly synthesised ligands at lower concentrations which are closer to those required for cellular assays.

Collaborator Contribution Dr Kyle Quinn, Bioengineering, University of Arkansas. This collaboration was to evaluate the effect of sAXL in an animal model of wound healing. Wound healing experiments were conducted in the Quinn lab, and the data was sent to the Higgins lab for further analysis. Dr Rodrigo Ledesma Amaro, Bioengineering, Imperial College London. In this collaboration, The Ledesma-Amaro lab is using a yeast toolkit to engineer recombinant sAXL for incorporation into a cellulose bandage.

Wound healing experiments were conducted in the Quinn lab, and the data was sent to the Higgins lab for further analysis. Dr Rodrigo Ledesma Amaro, Bioengineering, Imperial College London. In this collaboration, The Ledesma-Amaro lab is using a yeast toolkit to engineer recombinant sAXL for incorporation into a cellulose bandage.

PI Contribution The team from Imperial College London contributes in the development of the vessel detection and reconstruction method and subsequent avoidance strategy using Programmable Bevel-tip Steerable Needle system. The method will be based on the measurements of a laser Doppler flowmetry (LDF) monitor that is developed by Oxford Optronix Ltd.
Collaborator Contribution Since no commercial LDF monitor is suitable for the purpose of the SHINE project, Oxford Optronix is providing an upgraded version of their commercial, four-channel LDF system. This upgrade includes the ability to 'gate' reading from each channel, increased dynamic range in support of higher blood flow velocity, and a new type of LDF probe.

Description A laser Doppler flowmetry sensor is embedded within each segment of a programmable bevel-tip needle to detect a blood vessel. A type of machine learning network was used to interpret sensor data. Using this method, information with regards to vessel size and pose can be extracted.

Results and Impact • Hearing Futures II (in collaboration with the V&A Museum). This was an event organised to explore the need for additional research in the area of hearing assistive devices and music listening. This event fostered further networking activities, which brough to a further workshop held at Imperial College in January 2019, and consequent work for funding applications (EPSRC - Music and Hearing Impairment)

Description ICiC 6 - Rodriguez - The Great Exhibition Road Festival

Results and Impact During the Great Exhibition Road Festival, we presented a simulation of a steerable needle with a vessel avoidance system. In the simulation, audiences controlled the movement of the needle with a joystick, which emulates a pc game. This format of presentation attracted many children to try. After they played the 'game', we then explained how the system works.
