Molecular mechanisms of mRNA polyadenylation and deadenylation

Abstract

Our genes encode the information required to make the proteins in our cells. Genes are first transcribed into temporary copies called messenger RNAs (mRNAs). The mRNAs are then translated into proteins. Almost all mRNAs in our cells have a tail at one end – a polyA tail – that is important for controlling the stability of mRNAs and how often they are translated into proteins. The goal of my laboratory is to understand how polyA tails are added and removed from mRNAs.
We purify the cellular machinery that adds and removes polyA tails and then reconstitute their biochemical functions to understand how they work and how they are regulated. We use other methods, including electron microscopy, to determine their three-dimensional structures, allowing us to ‘see’ mechanistic details.
Addition and removal of mRNA polyA tails are fundamental processes, essential for controlling how genes are expressed. Therefore, it is important to dissect their mechanisms. Deregulation of polyA tails is associated with human diseases (e.g. cancer, heart disease, viral infection) and we hope that our work will provide insight into causes and potential therapeutic targets.

Technical Summary

Gene expression is tightly regulated to allow rapid responses to cellular stimuli. This is controlled through transcription, mRNA stability and the efficiency of translation into protein. Many of the post-transcriptional mechanisms of gene regulation are mediated by polyadenosine (polyA) tails which are found at the 3´-end of almost every eukaryotic mRNA. PolyA tails act in many diverse processes including the expression of genes in the inflammatory response, microRNA-targeted gene silencing and expression of maternal mRNAs in oocyte development. They are deregulated in human disease including cancer and viral infection.
My laboratory aims to understand the molecular mechanisms of addition and removal of mRNA polyA tails, thereby providing fundamental insight into the regulation of gene expression. We use an integrated, multidisciplinary approach combining structural, biochemical, biophysical, and functional techniques. We are determining high-resolution structures of Cleavage and Polyadenylation Factor (CPF), Ccr4–Not and Pan2–Pan3 protein complexes to elucidate how they determine polyA tail position and length. We will define fundamental principles underlying their assembly, gain insight into their activities and regulation, and establish the functions of individual subunits.
Specifically, we purify endogenous protein complexes as well as overexpressed (sub)complexes, to determine their structures and characterise their functions. We use electron cryo-microscopy (cryo-EM), alongside x-ray crystallographic, biophysical, biochemical and genetic techniques. We have developed methods for cryo-EM by designing new substrates that improve image quality, enabling structure determination of more difficult proteins than was previously possible.
An understanding of the structures and functions of CPF, Pan2–Pan3 and Ccr4-Not will provide fundamental biological insights into this exciting field.

Description The LMB appointed 30 Bird and public works as LMB Artists-in-Residence in 2015. They have been working with members of the LMB to inspire new perspectives and understanding of the LMB's research, both amongst people at the LMB, and the general public. As part of the Cambridge Festival of Ideas the Artists created an immersive experience piece: Home in The Service of Science. The events, which took place on 28, 29 & 30 October, were a great success with over 175 attendees learning more about the research conducted at LMB through conversations with scientists who performed in this unusual art-meets-science event.

Title ELECTRON MICROSCOPY SAMPLE SUPPORT COMPRISING POROUS METAL FOIL 
Description An electron microscopy sample support comprises: a support member; and a metal foil comprising a porous region. The support member is configured to give structural stability to the metal foil, and the porous region of the metal foil is configured to receive an electron microscopy sample. Also provided is a method of manufacturing such an electron microscopy sample support, a method of imaging using such an electron microscopy sample support and an apparatus operable to perform such imaging. An electron microscopy specimen support in accordance with aspects and embodiments may reduce particle motion and/or sample charging in electron microscopy, and thus improve information content available from electron micrographs. Appropriately designed and constructed supports may lead to an increased resolution per particle and increased accuracy of angular assignments in 3D reconstructions of, for example, biological specimens. This may enable the determination of structures of smaller and more difficult proteins than was previously possible using EM techniques.

Results and Impact The "Secret Universe" explained the inner workings of the human cell system, in a new, popular science programme screened on BBC2 at 8pm on Sunday 22 October 2012. The programme is also due to be shown on the Discovery Channel on Sunday 4 November 2012.

'Secret Universe: The Hidden Life of the Cell' used a range of interviews with experts and dramatic computer animations to explain what happens when a viral infection takes hold and how the body works to combat infection.

Results and Impact The LMB appointed 30 Bird and public works as LMB Artists-in-Residence in 2015. They have been working with members of the LMB to inspire new perspectives and understanding of the LMB's research, both amongst people at the LMB, and the general public.

As part of the Cambridge Festival of Ideas the Artists created an immersive experience piece: Home in The Service of Science. The events, which took place on 28, 29 & 30 October, were a great success with over 175 attendees learning more about the research conducted at LMB through conversations with scientists who performed in this unusual art-meets-science event.

Results and Impact Over 2000 visitors, including more than 600 children, took the opportunity to explore the work undertaken at the LMB, when it opened its doors to the public, on Saturday 22 June, to celebrate the opening of its new building and the centenary of the Medical Research Council (MRC). The family-friendly event showcased our work through lectures, tours, displays and hands-on activities for all ages.

this event was a great success with many people asking when we would do this again. It raised the profile of LMB in Cambridge

Results and Impact As part of its work in promoting careers in research to young people, Research Councils UK (RCUK) has commissioned Isinglass Consultancy to produce short case studies of funded researchers, to be published on the RCUK website.
