Biomarkers for nutritional intake and health

Abstract

Biomarker is a general term for substances, which are indicative of particular physiological states or processes. Their presence may show the risk or susceptibility for a disease, or even the progression of the disease itself. They can also be used to measure physiological processes such as energy expenditure. In clinical environments, biomarkers are often used for monitoring the success of a medical treatment, and similarly, they can show the effects of nutrient intake on nutritional status and health. ||At HNR we develop innovative technologies to measure such biomarkers, for processes such as insulin resistance, energy expenditure and the availability of dietary nutrients within the body. We conduct research aimed at identifying biomarkers for diagnosing and detecting the progress of nutrition-related human diseases e.g. obesity and diabetes, using modern biochemical and mass spectrometry techniques. When looking at the movement of dietary nutrients through metabolic pathways, we use a combination of techniques which use modelling of physiological systems i.e. breast feeding or gastric emptying, in conjunction with tracers techniques which incorporate stable isotopes. ||Scientists also perform research in the promising new field of nutritional metabolomics. This looks at the differences in the outputs from different metabolic pathways following different diets to increase our knowledge of human health and the interactions of diet and metabolism.

Technical Summary

The objective of the Bioanalytical Sciences section is to develop new and improved quantitative analytical methodologies for the discovery and interpretation of biomarkers of human physiology. The use of these techniques provides insights into the interrelationships of diet, nutritional status, and health, from which risk factors for disease can be identified, and nutritional strategies for its prevention developed. The research is currently focused in three areas: Biomarkers and Lipidomics. We are interested in the discovery and measurement of biomarkers for nutrition related diseases, using novel analytical technologies. We measure fingerprints of the entire lipid or content and its relative proportions. Biomarkers are then derived using multivariate metabolomics techniques, by observing significant correlations between pathology/phenotype and metabolite level (e.g. healthy vs. sick). We are also exploring the determination of multivariate profiles of pre-disease biomarkers, the study of which are particularly important to assess health risks. Protein prenylation. Isoprenoids may be functional biomarkers of diseases such as cardiovascular disease, cancer and neurodegenerative disease. HMG-CoA reductase is a critical element in the process of prenylation, where proteins are activated by attachment of isoprenoids. Isoprenoids are derivatives of mevalonate, which is the precursor of cholesterol. Recent evidence in cardiovascular and neurodegenerative research suggests that protective effects of certain nutrients (e.g. n-3 fatty acids) and drugs (statins) are due in part to a reduction of isoprenoid levels, in addition to reducing cholesterol. There is scant information on circulating levels of isoprenoids, however, in contrast to cholesterol. To better understand the nutritional regulation of HMG-CoA reductase as a key factor in protein prenylation and subsequent health benefits, we are studying isoprenoid levels under different treatment conditions, initially focussing on the impact of n-3 fatty acids and caloric restriction. Physiological modelling and isotope tracer techniques. The concentration of nutritionally relevant biomarkers in tissue is governed by the balance of the relative rates of formation and disposal or breakdown of metabolites. Physiological modelling is essential to understand metabolic processes and then test strategies designed to influence diet and health outcomes. Stable isotope tracer techniques enhance understanding of these models by allowing specific pathways to be studied, such as water fluxes (total energy expenditure, breast-milk intake), gastrointestinal function (gastric emptying and lipid absorption), lipid and glucose metabolism (insulin secretion and sensitivity, body composition) and nitric oxide production. They provide tools for measuring the benefits of nutritional strategies and allow optimisation of nutritional status in populations worldwide.

PI Contribution When mechanisms for lipid storage are compromised, as in severe insulin resistance (SIR), dietary fat may exert an increased influence on the balance of macronutrients oxidised. This effect on the macronutrient oxidation system maybe comparable to that of carbohydrate in healthy individuals. We hypothesise that in SIR, positive fat balance may induce an increase in the rate of total and the proportion of recently ingested fat oxidised. We plan to study this using a combination of stable isotope tracer methods together with indirect calorimetry. Furthermore advances in magnetic resonance spectroscopy will allow us to explore the role of liver in buffering recently ingested fat in SIR, particularly SIR associated with lipodystophy.
