Abstract

The Enabling Technologies programme consists of a number of core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. Because metabolic disease affects the entire body, it is vital to study these diseases whole animals. Our model of choice is the mouse. The Disease Model Core provides facilities for the measurement of the amount of fat and muscle in an animal’s body, how much and what type of food is eaten, and how much energy is burnt, through either metabolism, heat production, or exercise. It also provides facilities for examining isolated cells, tissues and organs. At the other end of the size spectrum, the Genomics/Transcriptomics Core enables scientists to study the sub-microscopic world of genes and how they function and are turned into proteins and the Core Biochemistry Assay Lab measures hormones and other molecules in blood.

Technical Summary

This programme provides the core facilities and technologies that act as vital supporting infrastructure, not only for MDU Programmes 1-4 but also for the other investigators across the MRL. At the heart of this programme are the Disease Model Core (DMC), the Genomics and Transcriptomics Core (GTC) and the Biochemistry Assay Lab (CBAL).

The DMC integrates two major platforms, physiology and histology, and provides a full range of support services to maximise scientific output from in vivo model systems. These include Home Office licensing support, cutting-edge in vivo phenotyping techniques, ex vivo tissue analysis, statistical analysis and data interpretation. The GTC provides access to high-throughput sequencing technologies, RNA quantification and ‘Sanger’ DNA sequencing as well as bioinformatics support. CBAL runs alongside the Clinical Biochemistry Lab of the Cambridge NIHR Cambridge Biomedical Research Centre and offers biochemical and immunoassay analyses (mainly rodent plasma and serum) to members of the MDU and IMS-MRL.

While the DMC and GTC are the primary facilities within Programme 5, a number of other key technologies, which receive the majority of their funding from other sources (Wellcome Trust and NIHR BRC), also provide critical support to MDU scientists. These are Microscopy and Imaging, Metabolomics/Lipidomics, Proteomics and the Genome Engineering Core. We seek to build on the success of these cores facilities and strengthen key areas where there is pressing, multi-user need.

Title High-throughput single cell unbiased lipidomics assay in human iPSC-derived dopamine neurons (AK) 
Description The development of single cell lipidomics approaches to study the lipid metabolism of dopaminergic neurons in relation to parkinson's disease. Changes in lipid metabolism have been strongly associated with Parkinson's disease (PD). Lipids have the potential to be important biomarkers for PD, but the lack of appropriate tools to measure total lipids (lipidome) in human dopamine neurons has limited advances in this field. we have developed a novel unbiased and high-throughput single cell assay to determine total lipids. This will allow us to identify and validate lipid metabolites in dopamine neurons, which may serve as biomarkers for PD.

Title bioinformatic pipeline for analysis of sequencing data from gene-edited cell lines (FM) 
Description CRISPR is a powerful way to edit stem cell clones, but identifying cell clones carrying desired edits remains challenging. To address this issue we developed GenEditID, a flexible, open-access platform for sample tracking, analysis and integration of multiplexed deep sequencing and proteomic data, and intuitive plate-based data visualisation to facilitate gene edited clone identification.

Results and Impact IMS staff and students worked with a local secondary school teacher and a communications trainer to develop 1 existing and 2 new activities for use with Year 9 pupils. We ran a whole day of activities on a bus with each of 2 schools considered to have 'under-served' intake - North Cambridge Academy and Ely College - meeting the complete Year groups in each case. We reached over 300 teenagers with the activities.

Feedback from our researcher volunteers:
- [Teachers] were also very complimentary and engaged with our activity as it touched on their lessons or personal lives
- I was surprised at just how smoothly it went, and by how engaged the pupils were Several teachers commented that they really liked the fact that we had three different activities that looked at different areas of research, but also themselves appealed to different personalities
- I was surprised how children very suddenly came up with very interesting and smart questionsit definitely makes you think about the scientific content in a different way!
- I learnt that secondary school children are being taught more complex science than I expected

Post-event responses from 62 year 9 pupils at North Cambridge Academy showed that
- 87% learned something new
- 79% did not have any relative or friend who worked in science or for a university
- just 11% (7 students) remembered ever going to the Cambridge Science Festival.
- 25% said that after the session they were more likely to consider a career in science.

Feedback on leaving the Institute from a staff member involved in development and delivery of this activity:
- Taking part in PE helped me to decide about the job change. I got the new job partly because of the skills and experience from being involved

While white fat stores calories, brown fat burns energy and could help us lose weight. Now, scientists have found a way of making the white fat 'browner' and increasing the efficiency of brown fat. While their study was carried out in mice, they hope that this finding will translate into humans and provide a potential new drug to help fight obesity.
