Abstract

The Enabling Technologies programme consists of two core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. Because metabolic disease affects the entire body, it is critical that one has model organism on which the proper study of the disease can occur. Our model of choice is the mouse. The Disease Model Core will provide facilities for the measurement of the amount of fat and muscle is in the body, and how much and what type of food that is eaten. It also has the capability to measure how much energy is burnt, either through metabolism, released as heat, or through excercise. It will also provide facilities for examining tissues and organs. On the other end of the size spectrum we have the other core facility that is part of this programme, the Genomics/Transcriptomics Core. As the disease model core studies whole organisms, this core provides the facilities to study the sub-microscopic world genes and how they function and are turned into proteins.

Technical Summary

The Enabling Technologies programme consists of two core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. The Disease Model Core will provide core facilities for the evaluation of body composition, food intake, energy expenditure and intermediary metabolism in rodent models. It will also provide facilities for histological processing and analysis. Importantly, it will have close links with the core phenotyping facilities associated with the Behavioural and clinical Neuroscience Institute so that opportunities for collaborative work are enhanced. The Disease Model Core also encompasses the Core Biochemical Assay Lab (CBAL). Although this is a primarily NIHR-supported core facility supporting all of the NHR Cambridge Biomedical Research Centre to undertake work that is strictly human based, funding provided through MRC MDU will continue to allow the CBAL to develop and deliver assays for analytes in mouse and rat specifically driven by the programmes of the MRC MDU and the wider IMS. The Genomics/Transcriptomics Core provides, and will continue to provide, facilities for DNA sequencing, mRNA quantitation and transcriptomic analysis, and which are local to our investigators and capable of delivering rapid turnaround for small- to mediumscale projects. This is highly complementary to the Eastern Sequence and Informatics Hub (EASIH), with which we maintain a close affiliation, and with the Sanger Institute.

Results and Impact Professor Fiona Gribble and Dr Giles Yeo (me) from the MRL and MRC Metabolic Diseases Unit in Cambridge, together with colleagues Professors Susan Jebb and Paul Aveyard from Oxford, featured in a 3-part BBC2 Horizon special 'What is the right diet for you?' in Jan 2015, following the trials and tribulations of 75 overweight volunteers looking to lose weight.

The programme put forward the key message that although obesity results from eating too much, the underlying influences on feeding behaviour are varied and complex, and explored whether diets could be 'tailored' to tackle these different factors.

In this instance, the investigators focussed on three groups of individuals - those with a higher genetic risk of becoming obese, those with a low gut hormone rise 30mins after a meal, and those who eat in response to stress.

By harnessing the power of genetics, together with biochemical and behavioural phenotyping, the programme provided a thoughtful and entertaining perspective on the current problem of obesity, and explored whether 'personalised' diets tailored to some of the root causes of overeating can succeed where more general dieting advice has failed.
