Abstract

The Enabling Technologies programme consists of two core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. Because metabolic disease affects the entire body, it is critical that one has model organism on which the proper study of the disease can occur. Our model of choice is the mouse. The Disease Model Core will provide facilities for the measurement of the amount of fat and muscle is in the body, and how much and what type of food that is eaten. It also has the capability to measure how much energy is burnt, either through metabolism, released as heat, or through excercise. It will also provide facilities for examining tissues and organs. On the other end of the size spectrum we have the other core facility that is part of this programme, the Genomics/Transcriptomics Core. As the disease model core studies whole organisms, this core provides the facilities to study the sub-microscopic world genes and how they function and are turned into proteins.

Technical Summary

The Enabling Technologies programme consists of two core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. The Disease Model Core will provide core facilities for the evaluation of body composition, food intake, energy expenditure and intermediary metabolism in rodent models. It will also provide facilities for histological processing and analysis. Importantly, it will have close links with the core phenotyping facilities associated with the Behavioural and clinical Neuroscience Institute so that opportunities for collaborative work are enhanced. The Disease Model Core also encompasses the Core Biochemical Assay Lab (CBAL). Although this is a primarily NIHR-supported core facility supporting all of the NHR Cambridge Biomedical Research Centre to undertake work that is strictly human based, funding provided through MRC MDU will continue to allow the CBAL to develop and deliver assays for analytes in mouse and rat specifically driven by the programmes of the MRC MDU and the wider IMS. The Genomics/Transcriptomics Core provides, and will continue to provide, facilities for DNA sequencing, mRNA quantitation and transcriptomic analysis, and which are local to our investigators and capable of delivering rapid turnaround for small- to mediumscale projects. This is highly complementary to the Eastern Sequence and Informatics Hub (EASIH), with which we maintain a close affiliation, and with the Sanger Institute.

Description We used liquid chromatography tandem mass spectroscopy (LC-MS/MS) to develop quantitative methods to analyse neuropeptide production in cultured human cells and in primary human brain samples. This consists of both standard curved made with sythetic human peptides, and synthetic peptides generated to incorporate stable heavy isotopes, which are spiked into samples during preparation. This enables very accurate and sensitive quantification of peptides, and assessment of relative quantities of different peptides of interests.

Impact This method enabled us to examine the relative production of POMC-derived peptides in both human cellular culture and in the primary brain. We found that alpha-MSH, which is the predominant MSH form studied, is produced at most lower levels than desacetyl alpha-MSH. Furthermore, we confirmed that beta-MSH, whose existence has been controversial, is indeed produced both in vivo and in vitro at similar quantities as desacetyl alpha-MSH.

Results and Impact Type 2 diabetes and obesity are life threatening metabolic diseases reaching epidemic proportions. The molecular basis for these diseases is incompletely understood, but genetic and environmental variables, synergistically interact throughout our lifespan to modulate metabolism. Today, there is growing appreciation that aging-related alterations in metabolism can be attenuated by lifestyle intervention strategies. Thus, insight into specific genetic, epigenetic, and cellular mechanisms that modulate metabolism throughout the aging continuum may uncover novel therapeutic entry points to manage type 2 diabetes and prevent obesity.

Results and Impact The heart of this programme was the effort to get a city in the UK, Newcastle, to lose 100,000 pounds. But clearly all the lessons learnt are broadly applicable across the UK and internationally. It was a three part series and I had a role in all three episodes. The first episode had me helping Hugh place the obesity problem into context. But it was the following two episodes which saw me going to Newcastle to try and engender real change on the ground. First I was given access to the central Newcastle metro station, where I placed footsteps leading from the platform and up the stairs, in order to try and 'nudge' commuters to use the stairs instead of the escalators. In the short-term, we did manage to get significantly more people using the stairs. Then I was given access to a street in Newcastle with a number of takeaway establishments. Here I manage to get them to swap out 'full-sugar' ketchup for the low sugar option (after a taste test to demonstrate that no one would be able to tell the difference), play about with portion sizes, as well as move full sugar beverages away from eye and grab height. What happened after the cameras left? Well, I went back to give a talk in Newcastle end of 2018 (the programmes had aired May 2018), I found that those footsteps that I had only put up at the central Metro station had now spread all across the city! In other stations, as well as in council owned buildings.

He is also an endocrinologist and an authority on the molecular causes of obesity and diabetes and previously served as Harvard Medical School Faculty Dean for Academic Programs and Chief Academic Officer for Beth Israel Deaconess Medical Center, a Harvard teaching affiliate. He is one of the country's leading investigators in the areas of obesity and diabetes. His research has produced major insights into the molecular mechanism of insulin action, the molecular mechanisms of insulin resistance in human disease, and the molecular pathophysiology of obesity.

Summary:Our bodies contain two types of fat: white fat and brown fat. While white fat stores calories, brown fat burns energy and could help us lose weight. Now, scientists have found a way of making the white fat 'browner' and increasing the efficiency of brown fat. While their study was carried out in mice, they hope that this finding will translate into humans and provide a potential new drug to help fight obesity.
