Protection of neurons in vitro and in vivo from Synuclein toxicity by molecular tweezers

Abstract
Parkinson's disease is an age-related neurodegenerative disorder in which aggregated clumps of a protein termed alpha-synuclein form in neurons, the cells in the brain which relay electrical information. These clumps are the defining key hallmark pathology of Parkinson's and are called Lewy bodies (LB). In a related disease, multiple system atrophy (MSA), the protein clumps form in the support cells, called glia. The diseases are therefore related and may benefit from a therapy to prevent the formation and spread of alpha-synuclein protein clumps.
The development of therapeutic strategies to prevent the formation and spread of alpha-synuclein protein clumps and prevent cell death in Parkinson's and MSA has been limited by a lack of understanding of the mechanisms driving neurodegeneration. However, increasing evidence demonstrates the importance of the alpha-synuclein clumps in disease pathology and has led to consider the therapeutic potential of several strategies aimed at reducing the burden and toxicity of alpha-synuclein accumulation. Our objective here is to build upon the unique experimental models of disease and complementary expertise of the Consortium. Members of the research team specialise in working in brain cells from mice, and with human neurons derived from stem cells made from Parkinson's patients. Others members of the team work on animal models of Parkinson's and MSA which model how alpha-synuclein may spread in the brain. We will use these models in laboratories in France and in the UK to test a novel molecular therapy called "molecular tweezer" drug which has been shown to inhibit aggregation and toxicity of other similar proteins which clump in other diseases. The drug may therefore protect neurons grown in the laboratory and neurons in the brain from Parkinson's or MSA-associated cell death. By probing the mechanisms and application of "molecular tweezers" to Parkinson's and MSA the project will demonstrate the use or not of such a therapy to prevent alpha-synuclein-mediated cell death.

Technical Summary
Parkinson disease (PD) is hallmarked by the deposition of aggregated alpha-synuclein (alpha-syn) species into intra-neuronal Lewy bodies (LB) and Lewy neurites (LN) inclusions while in multiple system atrophy (MSA) alpha-syn aggregates form oligodendroglial inclusions. The development of therapeutic strategies to block cell death in PD and MSA has been limited by a lack of understanding of the mechanisms driving neurodegeneration. However, increasing evidence of multiple pivotal roles of alpha-syn in the pathogenesis of PD and MSA has led to consider the therapeutic potential of several strategies aimed at reducing alpha-synuclein toxicity. Our objective is to build upon the unique experimental cell (Primary rodent neurons and glial cells - Human neurons derived from PD patients) and animal models of PD (SNCAOVX transgenic mouse and human PD-derived alpha-syn seed-injected mouse) and MSA (PLP-SYN transgenic mouse and human MSA-derived alpha-syn seed-injected mouse), developed by this consortium, for testing whether the prototypical molecular tweezer CLR01, shown to inhibit aggregation and toxicity of other amyloidogenic proteins, could protect neurons in vitro and in vivo from PD / MSA-associated synaptotoxicity and reduce PD and MSA's disease-like alpha-syn pathology. The project will thus (in)validate the molecular tweezers CLR01 for PD and/or MSA indications.
Planned Impact
The project will foster the development of disease-modifying treatment. To this end, it will develop and characterize a novel innovative molecule (CLR01) in clinically relevant human patient-derived dopaminergic neurons and in novel animal models to study its ability to interfere with alpha-syn pathology that it is induced by increased expression or by pathophysiological spread of patient-derived alpha-syn assemblies.
The synergy between the three complementary laboratories will maximize the potential for achieving these goals. We will efficiently share and transfer key materials, protocols and data between our laboratories, raising the chances of yielding synergistic, interdisciplinary, and groundbreaking research results. Hence, the outcome will be more than the arithmetic sum of each of our respective expertise, working force and the respective local academic environments. The multidisciplinary aspect of the project is essential to its objectives. It will benefit from expertise in many disciplines within neuroscience, delineating a clear synergistic network of competencies. Not only are different complementary capabilities needed to succeed, but a truly synergistic framework is mandatory. Our newly established links will allow implementing an innovative way of synergic work. Our contribution: we designed a cellular model of ?-synuclein propagation composed of dopaminergic neurons derived from healthy control iPSCs co-cultured with dopaminergic neurons derived from Parkinson's disease (PD) iPSCs harbouring a triplication of the ?-synuclein locus. I performed the cell culture, isolated the viable dopaminergic neurons and provided them to the group of Caleb Webber for single cell RNA-sequencing. |

Parkin is an ubiquitin E3 ligase that participates in several cellular functions, including mitochondrial homeostasis. However, the specific metabolomic changes caused by parkin depletion remain unknown. Here, we used isogenic human induced pluripotent stem cells (iPSCs) with and without PARK2 knockout (KO) to investigate the effect of parkin loss of function by comparative metabolomics supplemented with ultrastructural and functional analyses. PARK2 KO neurons displayed increased tricarboxylic acid (TCA) cycle activity, perturbed mitochondrial ultrastructure, ATP depletion, and dysregulation of glycolysis and carnitine metabolism. These perturbations were combined with increased oxidative stress and a decreased anti-oxidative response. Key findings for PARK2 KO cells were confirmed using patient-specific iPSC-derived neurons. Overall, our data describe a unique metabolomic profile associated with parkin dysfunction and show that combining metabolomics with an iPSC-derived dopaminergic neuronal model of PD is a valuable approach to obtain novel insight into the disease pathogenesis. | Results and Impact | Due to the COVID-19 pandemic, we were unable to hold the 2020 Alzheimer's Research UK Thames Valley Network Dementia Open Morning. Over the summer presenters within the network centre who were due to talk at the March public event to give their talks online. The short talks from scientists were recorded and include a range of topics in dementia. They are available on YouTube. | Results and Impact | Dr Pavandeep Rai was a judge in an Beyond Boundaries 2020 competition; an outreach event to schools in Oxfordshire. The University of Oxford invited schools to participate in an art competition for Oxfordshire state school students to create art inspired by research from Black, Asian and Minority Ethnic scientists. Students were asked to choose one of six scientists to create an artistic response to their research (e.g. painting, drawing, sculpture, animation, other artistic media) along with a short statement explaining the artwork. Due to COVID-19, there was a delay to announcing the winners. The winners are to be called in January 2021 and an exhibition of their work will take place online now. |

A video message was created from OPDC researchers thanking her for all the amazing work she has done towards funding Parkinson's research over the years. Together with a bouquet of flowers, they also gave her an engraved glass OPDC honorary award which was presented to her 'in recognition of outstanding fundraising to support research into Parkinson's'. |

| Results and Impact | On Tuesday 29th November 2022, Professor Richard Wade-Martins and Professor Michele Hu from the Oxford Parkinson's Disease Centre (OPDC) hosted the Oxford Parkinson's Research Day at the Kavli Institute for Nanoscience Discovery, Dorothy Crowfoot Hodgkin Building, South Parks Road, Oxford. Professor Irene Tracey, Vice-Chancellor Elect of the University of Oxford, Warden of Merton College and Professor of Anaesthetic Neuroscience, opened the days' proceedings. The programme was split across four sessions covering a wide range of translational work into Parkinson's: "Clinical Research into Parkinson's", "Translational models of Parkinson's", "Molecular Mechanisms and related biomarkers", and "Cellular models and target discovery" describing work across the University and from our collaborators. There were three invited keynote speakers:- Professor Masud Husain, University of Oxford. Masud Husain is Professor of Neurology & Cognitive Neuroscience at the University of Oxford. He leads the Neurological Conditions theme of the NIHR Oxford Biomedical Research Centre and is Editor-in-Chief of the publication, Brain. Together with Cornelia van Duijn, he leads Dementia Research Oxford. Professor Husain's presentation was titled, "When the spark goes out: the neurology of motivation and apathy". Professor Miratul Muqit, University of Dundee. Miratul Muqit is Professor (Clinical) MRC Protein Phosphorylation and Ubiquitylation Unit, School of Life Sciences, University of Dundee. His laboratory studies the protein kinase, PTEN-induced kinase 1 (PINK1) in which loss of function mutations cause Parkinson's disease. Professor Muqit presented, "Decoding Mitochondrial Damage Response Pathways linked to Parkinson's disease". Professor Sonia Gandhi, UCL. Sonia Gandhi is Professor of Neurology, Clinical and Movement Neurosciences at Queen Square Institute of Neurology, University College London. Professor Gandhi's laboratory works on the cellular and molecular biology of Parkinson's and her talk title was, "Thinking big to see small: from nanoscale to mesoscale resolution in Parkinson's". In addition to the talks, there were 37 poster presentations on display throughout the day. The event closed with Dr Kevin McFarthing, Research Advisor, Parkinson's UK Oxford Branch, presenting the best poster award voted for by all attendees. The prize awarded for the best poster was won by DPhil student Victor (Shijun) Yan, from the Molecular Neurodegeneration Research Group, University of Oxford, for his poster titled, 'a-synuclein in L1CAM-Positive Serum Extracellular Vesicles as a predictive biomarker for Parkinson's disease pathology'. | Results and Impact | Nora Bengoa-Vergniory poster "Tau-proximity ligation assay detects pathology ahead of tau neurofibrillary tangles in Alzheimer's disease" As the world population ages, neurodegenerative diseases are becoming the new epidemic in both developed and developing countries. Progress made in human genetics has revealed increasingly more disease-associated genes for Alzheimer's disease (AD), Parkinson's disease (PD), Frontotemoral dementia (FTD), amyotrophic lateral sclerosis (ALS) and other neurodegenerative disorders. These discoveries have linked neurodegenerative diseases with several common key biological processes that have so far been under-studied, including RNA metabolism, protein trafficking and innate immune responses. By focusing on these emerging areas in neurodegenerative disease research, this conference seeks to challenge the current research paradigms and to inspire in-depth discussion and exploration. The program assembles leaders from various fields to facilitate interactions between groups using diverse approaches, by providing a platform to cross-fertilize ideas and to encourage new collaborations that could lead to novel mechanisms and therapeutic targets against neurodegenerative diseases. |

| Results and Impact | On 19th June, a team of Unit members went to St Ebbe's C. of E. (Aided) Primary School in central Oxford, where they visited Year 6 pupils and their teachers to help them learn more about science, researchers, and how the brain works to control memory and movement. Pupils were first given a brief introduction to the work of the Medical Research Council and the MRC Brain Network Dynamics Unit. Pupils, teachers and Unit members then engaged in a range of interactive discussions and hands-on activities that included looking at nerve cells under a microscope, reporting on observations by making model cells, measuring electrical activity from muscles to control a robotic claw, comparing the brains of different vertebrates, discovering different types of memories, and using a game version of a brain-machine interface. |
