Nanoscale 3D architecture of signalling proteins in T lymphocytes

Abstract

The coordinated action of several protein kinases and phosphatases allows immune cells to
combat diseases and maintain health. To function, their organisation into micro- and nanosized
protein clusters appears integral. However, their coordinated spatial arrangement
during an immune response remains elusive. In this project we will employ state-of-the-art
highly multi-colour 3D super-resolution fluorescence microscopy, together with developing
new analytical tools, to address this question. We will use the developed approaches to map
the nanoscale organisation of key proteins during immune synapse development, in wild-type
and mutated cells carrying a genetic locus implicated with more than 16 autoimmune
diseases in humans
