Technical Summary
The interaction of proteins with immobilised ligands is increasingly important in areas like solid phase chemistry (biocatalysis) and screening of compound libraries on surfaces . However, there is little fundamental understanding of how the surface influences protein binding and catalysis. This study addresses two issues in biocatalysis on solid phase. 1) What causes the usually slow catalytic rates, and how may it be overcome? How important are factors besides simple accessibility of solid-phase substrates to the enzyme? 2) Effects on equilibrium position of reactions at a solid surface. Understanding gained will be directly applied to the enzymatic solid phase synthesis of dynamic combinatorial libraries to identify novel host-guest interactions (Joint with E20057).
