Towards a next-generation computational neuroscience

Abstract

What don't we know? Recently, the journal Science selected 25 of the biggest unanswered questions facing scientists over the next 25 years. Number two on the list, right after figuring out the composition of the universe, is: What is the biological basis of consciousness? This indeed is a big question. Scientific descriptions of conscious experience, volition, and subjectivity will follow in the footsteps of Copernicus and Darwin by restructuring our relationship with each other and with nature, and many clinical and technological applications will follow.A scientific account of consciousness will not arrive fully formed in a 'Eureka' moment. What is needed is a multidisciplinary, integrative approach combining theory and experiment and exploiting the interchange between the information/computation sciences and the neural, psychological, and medical sciences. At the front-line of this interchange, computational neuroscience (CN) uses computational approaches to model intricate brain processes in much the same way that meteorology uses computers to forecast the weather. In this view and in contrast to early approaches to 'artificial intelligence' (AI), brains are not computers, and intelligent behavior and conscious experience arise from complex brain-body-environment interactions unfolding in temporally precise ways. Much current CN focuses on single levels of description of neural systems (e.g., how neural activity affects connections among neurons) and neglects the multi-scale relations that connect brains, bodies, and behavior. Moreover, current CN is also surprisingly silent with regard to consciousness itself. By targeting and overcoming these limitations, our research will deliver new insights into the neural mechanisms underlying adaptive behavior and conscious experience. We will follow three interacting themes: (i) design and analysis of large-scale CN models to explore how multi-scale neural interactions shape and are shaped by brain-body-environment interactions; (ii) development of new theory to identify causal interactions in complex networks (what we call 'causal network analysis'), and (iii) creation of CN models that account for functionally significant aspects of consciousness, for example that each conscious experience integrates diverse information sources into unified scenes. Theoretical work in the above themes will interact with experimental data from multiple sources. At a fine-grained level we will characterize causal interactions in the intact brain of a pond snail, shedding light on the integrated neural function of a simple (non-conscious) organism as it interacts with its environment. Zooming out, we will apply causal network analysis to brain-imaging data acquired from humans in various states of consciousness, to test predictions based on CN models, and to guide the design of new models. Insights at the fine-grained level will scaffold our understanding of the more complex mechanisms underlying consciousness, with causal networks cross-cutting brains, bodies and environments providing a common theoretical framework. Taken together, these research strands will catalyze an important shift from correlation to explanation in consciousness science.As well as advances in basic science, our research will have important practical benefits at the interface of the biological and information sciences. These will include new design principles for AI/robotic devices, new insights for the design and control of complex technological networks, and new tools for the management of large-scale datasets. A next-generation CN will also underpin new clinical approaches. Many brain-related health problems, from coma to depression to insomnia, can be understood as expressions of disordered consciousness, and many existing clinical approaches are palliative and lacking in theoretical foundation. Our research will provide a theoretical basis for a new generation of effective clinical interventions.

Description This fellowship grant has supported research in both computational neuroscience and consciousness science. The focus of the research has been on connecting the two. What's important about this research? The biological basis of conscious experience is one of the outstanding challenges for 21st century science, and it is one that carries very important clinical implications as people suffer more-and-more from neurodegenerative and psychiatric conditions. Yet most of this research has so-far ignored the insights offered by advanced computational modelling. My research has contributed to closing this gap. With my team, I have delivered many new findings, insights, and tools. I have generated advanced methods for analysing causal interactions in neuroimaging data which make it possible to test new theories of consciousness, based on information integration, which we are now doing. I have developed some of these new theories, coupling the mathematics of Bayesian inference and information theory, with insights into conscious phenomenology. I have discovered how specific patterns of brain activity underlie conscious experience in particularly intriguing cases like synesthesia (where, for example, letters generate experiences of colour). Overall the work during the fellowship has led to the establishment of the world-leading 'Sackler Centre for Consciousness Science' which I co-direct. More than 100 papers have already been published by the Centre in the last 2-3 years.
Exploitation Route The findings guide ongoing and future research into understanding complex systems in terms of 'causal interactions', using the tools of information theory and time series analysis developed during the grant. The findings are also to substantive extent shaping the international research agenda in both basic and clinically applied consciousness science, leading to both new insights into biological mechanisms but also new approaches to diagnosis and prognosis in neurology and psychiatry.

Description Research during my fellowship has been instrumental in two dimensions. First it gave rise to, and continues to shape, the Sackler Centre for Consciousness Science, which I co-direct and co-founded. This Centre is now recognized as one of the, if not the, world-leading focus for research in consciousness science, and is uniquely interdiscplinary in bringing together mathematicians, physicists, and computer scientists with biologists and psychologists. This intramural dimension would not have been possible without the EPSRC fellowship. My work on consciousness science is having a large impact through public engagement in generating awareness of the how the brain and consciousness relate, both in health and in disease. This has really taken off lately, I have a TED talk with more than 7 million views which itself has very many ongoing impacts, and I am now an in-demand source of opinion on matters in neuroscience, AI, and consciousness - within academia and in many other areas, including e.g. film. Other specific findings have also found use. In particular, the work on time series analysis has led to a widely adopted MATLAB toolbox for analysing functional connectivity in time series data. This has been applied mainly in neuroscience but also in many other fields (economics, electrical engineering, etc).

Description There is no suitable choice in 'research tool'. We have developed a bespoke software toolbox which is now in its second iteration and is very widely used by multiple groups internationally to perform functional connectivity analysis. Results and Impact In Touch with Reality was an evening with the Sackler Centre for Consciousness Science, University of Sussex (led by Dr. David Schwartzman and Dr. Keisuke Suzuki) held at the Wellcome collection as an outreach event to the popular exhibition States of Mind. On the evening we attracted over 100 members of the public of all ages for this two hour event. The event has also inspired this article, "Cracking Consciousness" from James Hakner of the University of Sussex

Results and Impact We organized a unique 1-day public 'expo' at the Brighton Corn Exchange to provide a public dimension to our hosting of the ASSC2012 academic conference. This had never been done before (or since).

"State of Mind", ran for just a single day, was a free-entry event and attracted more than 1800 members of the public of all ages and all backgrounds. It was hosted by the Sackler Centre for Consciousness Science, University of Sussex as part of the first ever public outreach and engagement program in the 16 year history of the Annual Meeting of the Association for the Scientific Study of Consciousness (ASSC16), which this year was also hosted by the Sackler Centre.

State of Mind (www.consciousnessexpo.co.uk) was held in the Brighton Corn Exchange in order to explore and make accessible the exciting new science of consciousness. It was part of the Sackler Centre's public-facing events program towards making ASSC16 not just an academic event but a 'true citywide celebration of consciousness science'.

State of Mind showcased a fantastic range of new technologies and exhibits, developed by members of the Sackler Centre and colleagues from around the country, which exploited and explored the many aspects of consciousness, perception, and human experience.

The array of extraordinary exhibits drew from all aspects of consciousness science and included virtual reality environments, neuro-interactive films, mind-bending visual illusions, striking scientific images of the brain, eye tracking demonstrations, a Turing test, artificial intelligence, sensory substitution devices, impossible objects as well as real experiments in which visitors could take part in. Many of the exhibits had a strong interactive component, allowing visitors to be immersed in, and to learn about, different aspects of their own conscious experience. There were also many opportunities for visitors to talk with the scientists, technologists, and artists currently investigating the biology of consciousness, as well as roaming experts on consciousness - drawn from the Sackler Centre - that visitors could approach with their queries.

Results and Impact Primary scientific advisor to the major Wellcome Collection exhibition' "States of Mind" which ran from October 2015-2016 and was visited by 190,000 people. With my lab, we provided content, advised on content, co-wrote exhibition texts, gave 'expert' guided tours, designed bespoke iPad applications, wrote essays for the Wellcome website and organised a 1-day 'virtual reality' installation with 20 researchers and 15 different VR-based interactive experiences.

Results and Impact I was one of nine featured scientists in this major feature documentary by Vice/Motherboard, directed by Ian Cheney and with Werner Herzog as advisor. The film premiered at CPH:DOX, was screened in theatres internationally, and reviewed well in many media including The New York Times, LA Times. It is now streaming on Netflix. The documentary highlighted the work of the Sacker Centre for Consciousness Science to a global audience and its impact is still being measured. Initial communications from the general public have included further invitations to speak or be interviewed and conveyed a sense that this documentary made our research more accessible to a non-specialist audience.

Results and Impact This was a 2 day workshop to foster creative thinking within my research group. We engaged the services of a professional facilitator to ensure beneficial outcomes. The workshop was funded specifically by the EPSRC Creativity@home fund, which was available to fellowship holders.

We had many new ideas for collaborations within Sussex which are now being followed up.
