Manufacturing research hub for a sustainable future: Vaccines for pandemic preparedness

Abstract
The Covid-19 pandemic continues to take a huge toll - an estimated 6.3m people have died including 178,000 in the UK. Globally 1.6bn students have missed school, 250m people will be pushed into extreme poverty and economic losses are estimated at £12tr. History shows that epidemic and pandemic threats constantly emerge, whilst SARS-CoV-2 continues to mutate as it becomes endemic. It is clear that major losses could be prevented by sustained domestic investment in public health. Work undertaken within Vax-Hub1 on responsive technologies and accelerated quality control methods enabled rapid development and manufacture of the ChAdOx1 vectored vaccine against SARS-CoV-2 (licensed for emergency use in December 2020 via a non-profit partnership with AstraZeneca). Over 2.9bn doses have now been released in 180 countries. The UK had a leading role during the pandemic and the proposed Hub builds on this success to advance novel research on a broader range of technologies. Working closely with stakeholders, Vax-Hub will enable the UK to be better prepared for the next pandemic.
This investment into The Future Vaccine Manufacturing Hub will enable our vision to make the UK the global centre for vaccine discovery, development and manufacture. The Vaccine Manufacturing Hub brings together a world-class multidisciplinary team with decades of cumulative experience in all aspects of vaccine design and manufacturing research. This Hub will bring academia, industry, not-for-profit organizations and policy makers together to propose radical change in vaccine development and manufacturing technologies, building on a technological innovation culture.
The Hub will enhance future vaccine manufacturing through (i) de-risked manufacture of new vaccines by strategically innovating for a selected range of the most promising platform technologies (established and novel/disruptive); (ii) developing manufacturing options that improve the product quality and so immunogenicity; (iii) streamlined manufacturing process development with novel responsive solutions and advanced digitalisation strategies; (iii) a focus on enhancing stability and needle-free administration routes so they become a reality within the lifetime of the Hub. The proposed Hub would be the natural location for early-stage research before projects are transferred to a GMP manufacturing facility.
The work focuses on development of improved vaccine platforms which can be flexible enough to be used for multiple product manufacture. These improved vaccine technologies are used as case studies to test rapid and responsive development tools to create a whole process mimicking vaccine manufacture, which could be easily and quickly deployed in case of epidemic/pandemic scenario. Finally the research focuses on standard and novel adjuvants to make mucosal delivery a reality, thus allowing alternative route to injection for mass administration.
The Hub will establish the UK as the global centre for end-to-end vaccine research and manufacture. Additionally, vaccines should be considered a national security priority, as it is evident that diseases do not respect international boundaries, thus this work into capacity building and rapid response is a significant advantage. | Description | It is an installation for bioremediation, in particular the remediation of greywater and surface run-off for a future Biocene house. The structure is proposed as a means of decentralised remediation of a complex mix of surfactants, phosphates, heavy metals and other organic molecules. Previous work has shown that even challenging and toxic compounds can be degraded using consortia of algae and bacteria working in parnership and exchanging nutrients to form a stable autonomous system. Biofilms are emergent assemblages of algae and bacteria that have important roles in nutrient cycles and remediation of pollutants. Through computational design and advanced manufacturing we create a water recirculating mechanism that promotes the growth of microbial communities, enhancing the acceptability of such potentially abject conditions in heterogenous biomes. |

| Results and Impact | Sean Elias organised the pop up event 'Research for a Health Future' on 20 Feb 2025 as part of the Festival of Tomorrow in Swindon. This was a collaboration between researchers based in Oxford University and UCL with VaxHub a central part. Reshma Kailath attended in a supporting role. Their engagement activities are to help out with the future pandemic planning, winter viruses (with focus on flu), vaccine delivery routes (needle vs aerosol vs nasal etc), vaccine manufacturing and improving vaccine immunity. The aim was to establish Swindon as a new location for targeted engagement and forge links with local community groups. A major outcome was that Sean Elias set up a future collaboration with the NHS Bath and North East Somerset, Swindon and Wiltshire Integrated Care Board (ICB) to share information about the work of VaxHub with its members and local community. |

The event was an opportunity to hear first-hand about the Hub's vision to deliver manufacturing innovations that enable a transformative change towards sustainable and rapidly responsive vaccine manufacture. In addition to hearing about the new Hub, attendees also had the opportunity to learn how industry are tackling the challenges presented by the need for improved manufacturing sustainability with confirmed presentations from AstraZeneca, Croda and Synthace. The event included dynamic opportunities for networking with both academics and other industry partners. The feedback from the event was positive and has led to discussions with multiple organisations who attended and now also are interested in becoming members of the Hub. |

To achieve so, this project offers practical laboratory experience - complementary to taught material - which is not accessible within traditional school environments. On the 25th of January, Vax-Hub hosted Y12 students from Langdon Park School at UCL Biochemical Engineering. Students attended a masterclass on vaccine development and manufacture, followed by practical laboratory activities using SDS-PAGE, and a tour of the department lab facilities. |

From discovery science, through manufacture all the way to clinical trials and mass vaccination. People were even offered the opportunity to receive a flu vaccination there and then. Prior to COVID-19, my team designed vaccines against Ebola, Mers and influenza, and used the same vaccine platform technology to create our COVID-19 vaccine. The conversation also covered early career interests and motivations, the development of the vaccine during the pandemic, the field of vaccine research since COVID-19, and how we prepare for the challenges ahead. |

| Results and Impact | We hosted the VaxHub Futures Forum on Pandemic Preparedness during UCL's Festival of Engineering. The event brought together over 50 participants from academia, government, industry, funding and advisory bodies to showcase the hubs' vaccine manufacturing research and explore potential future scenarios for pandemic preparedness through the Futures Forum methodology, developed by Dr Ine Steenmans. The Futures Forum is an innovative and adaptable knowledge exchange format which uses 'futures thinking' to guide participants through a joint exploration of futures of a particular policy topic. Following the event, we published a report which compiles insights that emerged from the participants' discussions during the VaxHub Futures Forum, as they were asked to place themselves in the year 2030 and to reflect on their hopes, fears and the challenges associated with pandemic preparedness. The key themes stemming from discussions and highlighted in the report include equitable access to vaccine technologies and vaccine manufacturing capabilities; public health systems resilience; knowledge exchange, interdisciplinarity and collaboration; and public engagement and trust in science. |
