Understanding the differential and complementary roles of SGK isoforms associated with the PI 3kinase and Akt signalling

Abstract

This project work builds on previous exciting collaborative research between the Alessi lab and AstraZeneca that indicated that SGK protein kinases play critical roles in driving proliferation and survival of cancer cells (Sommer EM et al., Elevated SGK1 predicts resistance of breast cancer cells to Akt inhibitors, Biochem J. 2013;452:499-508 {http://www.biochemj.org/bj/452/0499/bj4520499.htm} and Bago, R et al., Characterization of VPS34-IN1, a selective inhibitor of Vps34, reveals that the phosphatidylinositol 3-phosphate-binding SGK3 protein kinase is a downstream target of class III phosphoinositide 3-kinase. The aim of this studentship will be to undertake research in order to define how SGK isoforms are regulated and how they function. A key question will be to determine whether inhibiting SGKs could result in a novel therapeutic strategy to better treat cancer in the future.
