Directed Protein-Protein Interactions Networks

Abstract

The first stage of this project will be to design a method to infer the best directed Protein-Protein Interactions (PPI) network based on available aggregated PPI (and related) data (ideally data obtained from Human Plasma). To achieve this, we will have to review the different databases available and assess their comprehensiveness and their reliability. We will also need to review what has been done until now in Networks analysis and more specifically in the study of directed networks (applied to PPI networks but also to genes regulatory networks for example). This first step will also be the time to study potential inter-species redundancies, as a way to enrich the Network, and to consider how to use genetics and Mendelian Randomization (MR) methods to help to direct the edges.
For the second step, we will try to corroborate the results obtained at the first stage using data from the INTERVAL study. We will test the different models developed previously and try to optimize the integration of the various tools (Networks analysis, MR etc.).
