Radio-labelled responsive fluorophores to elucidate the role of MMP in ageing

Abstract

Matrix metalloproteinases (MMPs) play an essential role in a variety of biological processes,
including ageing. Within this project, a new platform of multifunctional probes for the detailed
study of MMP's role in age-related diseases will be developed. The probes will consist of an
organic fluorophore covalently linked to a PET-tracer and anchored via an MMP cleavable
peptide sequence to a black-hole quencher (BHQ). The PET-tracer will allow for the spatial
monitoring of the probe's diffusion in the cellular matrix, while the BHQ will add temporal
resolution when cleaved by MMP, thereby "turning-on" the fluorescence.
