Novel 3D printing approaches to induce intestinal epithelium development from adult derived gut organoids

Abstract

This project will develop an in vitro 3D model of the normal, intestinal epithelium to achieve a cell-based model that more closely replicates intestinal epithelial structure and function as seen in vivo. Such a model will address the enormous variability associated with organoid culture which leads to issues regarding reproducibility between experiments. Our innovative approach of using 3D printing to produce 3D structures that mimic the geometry and cellular organisation as seen in vivo, will accurately reproduce the intestinal epithelium in vitro and ultimately support the translation of basic bioscience.

This inter-disciplinary research project therefore provides an exciting opportunity to redefine 3D culture of epithelial tissues using state-of-the-art bioprinting techniques. The student will utilise additive biofabrication, including additive manufacturing and microfluidics, that allow the formation of 3D cell culture substrates with defined architectures within dynamic culture conditions. In addition, the successful candidate will be able to develop skills in mammalian cell culture, including of primary cells, and in their characterisation following culture.
