Next Generation Analytical Tools: Application to Protein Oxidations that affect Human Health and Wellbeing

Abstract
New and novel measurement tools and technologies enable modern biological and biomedical research to continue to move forward at a rapid rate. Detecting disease and evaluating the effects of drugs all rely on measurement technologies, and the limitations in our ability to measure things rapidly, accurately and cheaply is often the bottleneck in many research areas. Similarly, advances in industrial processes and clinical methods are often dependent on matching advances in measurement technology. In this programme two leading research centres from Glasgow University and Imperial College London join forces to develop new techniques and tools in order to improve existing approaches to detect and quantify the effects of protein oxidative damage in biological samples. Protein oxidative damage is one of the components of ageing and can cause a range of serious medical problems. As part of this project we will improve the ways in which biological samples are delivered into a range of analytical platforms such as mass spectroscopy and protein microarrays by employing novel innovative approaches such as acoustic sound waves, emulsion droplets and microfludics. Chemical tools and probes that seek out and detect damaged proteins will be constructed and subsequently incorporated into the technologies mentioned above. These technologies will be used to study the dynamics of protein damage in space and time. Another part of the programme develops high throughput analysis tools of various forms all of which use minaturisation methods to allow for the analysis of as many proteins as possible, as rapidly as possible and with as much detail as possible. It is anticipated that this multidisciplinary programme will create the necessary analytical platforms to overcome a technological gap that is holding back biological and medical discoveries in this research area that is vital for fostering our understanding of human health and ageing. It also has the potential to facilitate medical interventions to alleviate the symptoms of ageing and age-related illness. Given the importance of these issues for the general population, not just of the UK but worldwide, the impact of this programme will be significant and sustained for several years in terms of academic output as well as industrial wealth generation. In particular, novel drug targets and indeed drugs, as well as improved diagnostic techniques to monitor and assess human health and ageing are possible outcomes in the near future. Given the importance of oxidative damage in crop production it is likely that our programme will also produce tools that benefit the agrochemical industry, making this proposal a truly discipline crossing venture with respect to the sciences involved and the impact generated.
Planned Impact
This project will be of benefit to both the academic and industrial communities and is aligned with three of the EPSRC's strategic areas (Nanoscience through engineering to application, Ageing-Lifelong Health and Wellbeing, and Towards Next Generation Healthcare). The programme which capitalises upon seven-state-of the-art technologies developed within the ICB DTC and C&PT DTCs will realise a number of key advances with broad-reaching application. It is directly focused on the development of tools that will enable drug discovery and disease proteomics, with improved healthcare as the long-term goal. The advances that will benefit both sectors include technical improvements in mass spectrometry, multidimensional spectroscopy, chemical probes, protein arrays, microfluidics and proteomic activity profiling. In addition, the tools and techniques under development here have the potential to provide enhanced capabilities for end-users in basic biology, biomarker discovery, diagnostics and drug discovery. These applications can be directly exploited by the pharmaceutical industry, medical sector, biotechnology SMEs and instrumentation manufacturers. The project builds on our existing doctoral training programmes and will provide training of a cohort of PDRAs and students in the important area of post-genomic technology and novel analytical tools. There remains an unmet need for highly capable multidisciplinary scientists with genuine expertise across more than one discipline. Our programme will produce future designers and developers of new measurement techniques and strategies via a cohort who understand both a range of technologies and the details of the biological and biomedical challenges to be addressed. The transferable skills we will provide will include courses on systems integration, entrepreneurship and exploitation, allowing them to translate and exploit their research both during the project and their future careers. The focus of the programme is concerned with providing tools for the better study of and intervention into oxidative damage of proteins, which is one of the molecular components of ageing. There are obvious benefits to the general population of research that has the potential to impact on the adverse effects of ageing and improve quality of life. UK industry will benefit from the scientific and technical innovations we will produce, with a 5-10 year timeframe for commercial realisation. The diagnostics tools that we will develop will not only be able to detect the changes associated with disease or ageing but also, and most importantly, to monitor their progress during the drug development process and subsequent therapy (10 yrs). The economic imperative is for medicine to be more preventative; these tools will play a vital role by facilitating the identification of illness in its early stages, allowing for early intervention to reduce permanent damage (10 yrs) and alleviating associated healthcare costs. In the context of drug discovery, the ability to understand and study the enzyme active sites and peripheral damage to easily oxidised signalling systems paves the way for novel intervention strategies either to mitigate aging effects or to target these proteins as part of an intervention for other diseases. Our technologies have the potential both to identify and to validate targets and biomarkers, as well as testing for side effects of drug candidates (5-10 yrs). Individual elements such as the MAC chips and 2DIR will be exploitable for clinical diagnostics and drug discovery on shorter time scales (ca. 3-5 yrs) and the proteomic technologies we will develop can be applied to any disease, including cancer, and cardiovascular diseases. The result will be improved healthcare as well as enhanced economic competitiveness of the United Kingdom by generating wealth and potential future employment.

Description | A humidity cell was made from a cheap optical mount, a CaF2 window, a glass coverslip, PTFE spacers and a rubber o-ring. The biological sample of interest is placed on the glass slide along with several drops of saturated salt solution, which enable a constant humidity to be maintained inside the cell for at least a month. This ensures that the water content of the biological sample of interest remains constant throughout a series of 2DIR experiments. |

This leads to higher throughput in our 2DIR experiments. |

Cells are loaded into these devices, transported to analysis chambers using an optical trap and then ruptured with the use of a high powered optical lysis laser. The intracellular contents are then quantified by the use of a miniaturized ELISA format. Proteins bind to antibodies on the surface of the device and a secondary fluorescent antibody, present in solution completes the sandwich system. Total Internal Reflection Fluorescence (TIRF) microscopy is then used to detect and count the amount of protein that has bound to the antibody spot. |

Description | A technique using a short pulsed laser is employed to lyse single cells. Through a series of physical processes a cavitation bubble expands from the focal point of the laser. The shearing forces experienced by the cell placed approximately 10_m away disrupts the cell and liberates intracellular constituents for analysis. It has been demonstrated that the cell may be fully (all membranes ruptured including nuclear membrane) or partially (only plasma membrane ruptured preserving the nucleus intact) lysed. Title | Acoustic suppression of the coffee-ring effect. |
Description | We study the influence of acoustic fields on the evaporative self-assembly of solute particles suspended inside sessile droplets of complex fluids. The self-assembly process often results in an undesirable ring-like heterogeneous residue, a phenomenon known as the coffee-ring effect. Here we show that this ring-like self-assembly can be controlled acoustically to form homogeneous disc-like or concentrated spot-like residues. The principle of our method lies in the formation of dynamic patterns of particles in acoustically excited droplets, which inhibits the evaporation-driven convective transport of particles towards the contact line. We elucidate the mechanisms of this pattern formation and also obtain conditions for the suppression of the coffee-ring effect. Our results provide a more general solution to suppress the coffee-ring effect without any physiochemical modification of the fluids, the particles or the surface, thus potentially useful in a broad range of industrial and analytical applications that require homogenous solute depositions. |

Results and Impact | Annual event that takes place at the Royal Society in London. Around 20 groups from around the UK and beyond are selected to take part in this prestigious event. The event involved constructing a stand and a series of interactive demos. This was split across the three institutions, Imperial, Glasgow and Aston with each institution creating one demo or activity. The event spanned an entire week which involved interacting with secondary school students, members of the general public and fellows of the Royal Society. Since the event we have a number of requests for more detailed information which related to our stand. This included an A-Level student asking for information about oxidative damage for a school presentation and a student at the Royal College of Art asking for information about how sound waves can be used to interact with biological material. |
