Description Expression of CYP1A protein form the Rainbow Trout has been achieved and a method developed for its isolation and purification. Structural analysis and characterization of the CYP1A protein has begun and data is being collated. Comparison with the Human counterpart is being carried out.
Expression has also been achieved and method developed for the truncated CPR enzyme and an efficient purification protocol has been optimised. The enzyme will be then used as an effective redox partner enzyme for the purified CYP1A protein and, ultimately, other P450 enzymes of interest. Structural analysis and characterization of the CPR protein has begun and data is being collated. Comparison with the Human counterpart is being carried out. Expression of CYP3A126 protein from the Fathead Minnow has been achieved and a method developed for its isolation and purification. Structural analysis and characterization of the CYP3A126 protein has begun and data is being collated. Comparison with the Human counterpart is being carried out. The cytochrome P450 enzymes substrate profiles are being collected, this will include a range of known toxins. Following this metabolites will be collect and analysed via mass spectrometry.
Exploitation Route The species chosen for this project represent a range of organisms which are already used as model organisms by research agencies, the government and industry. Cytochromes P450 can potentially be used as biochemical markers; since defining their function and mechanism of action (including their metabolites) on these known toxins they will provide an in vitro determination of harm and risk. The development and use of in vitro and predictive models to determine harm and risk of chemicals has increased recently, moving away from the traditional animal models as people become more aware of their impact on the environment and other species. The overarching aim of this project is to develop further understanding of important cytochrome P450 enzymes which may be conserved across species, Cytochrome P450 Enzymes from Environmentally Sensitive Organisms their function and the interaction of these enzymes with environmental toxins. The data gathered will provide deeper insights into the interactions of environmental toxins in particular, thus providing better environmental risk models for human activity.

Results and Impact 45 postgraduate PhD students attended the Innovation & Commercialisation of Research unit which I had been asked to help out. This involved me discussing my work history and discussing my experience of patents and intellectual property. This triggered questions and discussions as I circulated the room to aid them in their tasks. I was asked to return the following week to obsere their presentations and provide feedback.
