Abstract

Abstracts are not currently available in GtR for all funded research. This is normally because the abstract was not required at the time of proposal submission, but may be because it included sensitive information such as personal details.

Technical Summary

We will generate high quality transcriptome datasets with RNA-seq. These will provide two important sets of data. Pre-vaccination datasets will give insight into the overall genome-wide transcription output in PBMCs from different cohorts, which may uncover differences in overall immune competence due to environment/ethnic background. VDJ-seq datasets will provide a detailed picture of antibody diversity as a predictor of immune competence before vaccination and a marker of antibody expansion post-vaccination.

Description Response to a malaria vaccine in an African population are augmented by the use of an alternative adjuvant to the one currently used, both in terms of T cell and B cell activation and antigen receptor repertoires.
Exploitation Route It could lead to further studies on increasing effectiveness of vaccines.

We have published a manuscript to disseminate our findings (Hill et al J Exp Med 2019), which has been highly read and cited by researchers in a wide range of vaccine-related research areas.

PI Contribution Analysis by next generation sequencing of antibody repertoires of subjects vaccinated with a malaria vaccine with different conjugates to determine relative vaccine efficacy. This will provide a data resource comprising datasets of immune repertoires in a Tanzanian population as well as functional evaluation of vaccine efficacy.

Results and Impact 180 sixth form aged students attended this event where staff discussed and answered questions about what a bioscience career is like as well as discussing research interests. Teachers reported that students had increased their knowledge of such careers and especially highlighted the breadth of different roles the event highlighted to them.

Results and Impact A recorded Q&A discussion video made in collaboration with the British Society for Immunology (BSI) where public questions around the COVID-19 vaccine were submitted prior to the event and then answered. To date over 500 people have watched the recording published on the BSI social media platforms with audience feedback suggesting that it has led to a change of views / understanding of the science behind vaccination.

Results and Impact I gave a 1.5 hour lecture on B lymphocyte development, antibody repertoires and our studies on malaria vaccination to a group of 30 third year undergraduate and MSc students from the University of Makerere, Uganda by video link. The lecture was also filmed and put on their website. This was organised by Cambridge for Africa as part of a lecture series by immunologists associated with the University of Cambridge. There were numerous questions afterwards, and positive feedback from the students to the Cambridge for Africa organiser.
