Technical Summary
Cambridge University was awarded #665K to expand the Metabolic and Cardiovascular disease facility for the development of mouse models. This will add significantly to the existing strengths at Cambridge, and directly addressed the MRC strategic priority of developing models of human disease.|Metabolic and cardiovascular diseases are major public health challenges. Additionally, therapeutic options for these disorders are very limited and thus, they represent, to industry, a considerable unmet need. Cardio-metabolic diseases are quintessentially disorders of systems, not simply cells, and studies in whole organisms are therefore essential to both the elucidation of their pathophysiology and the examination of possible novel therapies. The pharmaceutical and biotechnology industries are increasingly looking to partner with sophisticated academic laboratories to undertake sophisticated target validation and proof of concept studies in appropriately designed and refined animal models. The UK academic sector, in general, is not particularly well placed to respond to this need. However, there are key centres of excellence in specific disease areas that are uniquely placed to fulfil those needs. Cambridge University may represent such a centre in cardio-metabolic disease and the award will helps to improve the Universitys ability to carry out detailed phenotypic analysis of murine models of human metabolic and cardiovascular disease. The development and utilization of sophisticated phenotyping technologies are crucial to capitalise on the power of murine genetic manipulation and facilitate meaningful translation of basic science into the clinical arena. Cambridge aim to incorporate state of the art technologies and develop sophisticated, accurate protocols in two major phenotyping areas: 1) the characterization of central and peripheral mechanisms controlling energy balance (e.g. food intake, energy expenditure and fat deposition); and 2) the evaluation of cardiovascular complications such as hypertension, dyslipidaemia and macrovascular disease that are strongly associated with metabolic diseases such as obesity and diabetes mellitus.

| Results and Impact | EAS Advanced in Cardiometabolic Phenotyping in Rodents, Cambridge, UK Scientists from the University of Cambridge Biomedical Campus recently hosted the 2nd EAS Advanced Course in Cardiometabolic Phenotyping in Rodents. This 3-day course combines the state of the art facilities and expertise within the Disease Model Core at the WellcomeTrust-MRC Institute of Metabolic Science and the BHF Cardiovascular Centre. By integrating the expertise of these two specialized phenotyping centres, it provides a unique opportunity to learn about many of the skills required for in depth cardiometabolic phenotyping in small animals. The 32 participants, selected from centres across Europe, were treated to a programme of lectures, phenotyping demonstrations and data analysis sessions. A further 1-day workshop at the Sanger/EBI focused on mouse genetics, genetic engineering and informatics resources, highlighting the great opportunities provided by recent advances in genetic engineering purpose-made mouse models. Participants particularly appreciated the mixture of hands-on experience, combined with opportunities to ask detailed questions on how the techniques demonstrated should be used to suit their own research priorities. The course was sponsored by the European Atherosclerosis Society, British Heart Foundation and TSE systems, with organisation led by Prof Toni Vidal-Puig, Director of the MRC Metabolic Diseases Unit Disease Model Core. |

Sam's talk was focussed on metabolomics in endocrinology and was based on the past 9 years of experience in the TVP lab in the field of lipodomics. The purpose of the talk was to discuss how biologists without systems biology experience can work with groups specialising in the fields of metabolomics to design appropriate experiments and extract the maximum value from their data sets. | Results and Impact | I'm a Scientist - Get Me Out of Here! During the lockdown, two members of the TVP Lab - Sonia & Jennie - took part in the MRC initiative 'I'm a Scientist - Stay at Home!'. This programme was established to allow school children to talk to real scientists during their virtual science lessons in the summer term, to find out more about the subjects they were interested in! Sonia was in the 'Disease Detectives Zone', while Jennie was in the 'Medical Research Zone'. They both had a lot of fun, and were amazed by how insightful the questions they received were! |

We have been approached by other academic institutions and companies to provide advice on metabolic phenotyping and to request technical support to develop their own programmes. Also we are increaisngly receiving request to review grants based on technology for murine phenotyping. |

These difficulties are causing mental health problems, frustration, anger, and disengagement. However, this turbulent time has also enabled new, unexpected leaders to emerge, performing more autonomously and making insightful decisions. This period has provided them with opportunities to show their character, determination and resilience when confronted with challenges. |
