Development of NanoKick Bioreactor

Abstract
Bone graft is the most transplanted tissue after blood. It is in short supply and demand is increasing with the aging population. Taking graft from a patient to transplant back to the patient is possible but results in morbidity of the donor site (normally the pelvis). We have developed a novel bioreactor, called Nanokick, which can be used to efficiently and effectively convert mesenchymal stem cells (MSCs - the bones natural stem cell supply) into osteoblasts (bone building cells). It works by supplying tiny kicks to cells. Our approach will provide technology that enables provision of 3D living bone derived from a patients own cells that produces effective repair. Areas this would be useful would include all bone reconstructions, spinal fusion and joint replacement. These in no need for chemicals and associated side effects (e.g. widely used bone morphogenic protein is now associated with adverse effects). In this work we will develop our technology towards a commercial product.

Description | We have developed a bioreactor that can be used to engineer 3D bone that could be used for drug testing instead of animals. The bioreactor could also be used to produce e.g. tissue engineered bone graft. As part of this project, new applications for the bioreactor have been identified through new collaborations. |

Description | The results are being used in outreach e.g. Royal Society Summer Exhibition 2016 and 2018. Also, we are building an impact case around our bone technologies of which nanokicking is one. Cells produced by the bioreactor will be used in a human trial in 2023 for bone repair. As part of this bioreactors will be provided to contract manufacturers to produce a cell therapy and have been placed into a GMP manufacturing facility who have started to manufacture cells for pre-clinical testing. We are now trying to licence the bioreactor. |

Description | Models of bone homeostasis could be useful in drug discovery, particularly if the model is one that replicates bone regeneration with reduction in osteoclast activity. Here we use nanovibrational stimulation to achieve this in a 3D co-culture of primary human osteoprogenitor and osteoclast progenitor cells. We show that 1000 Hz frequency, 40 nm amplitude vibration reduces osteoclast formation and activity in human mononuclear CD14+ve blood cells and enhances osteogenesis/reduces osteoclastogenesis in a co-culture of primary human bone marrow stromal cells and bone marrow hematopoietic cells. Further, we use metabolomics to identify Akt (protein kinase C) as a potential mediator. Akt is known to be involved in bone differentiation via transforming growth factor beta 1 (TGF?1) and bone morphogenetic protein 2 (BMP2) and it has been implicated in reduced osteoclast activity via Guanine nucleotide-binding protein subunit a13 (Ga13). With further validation, our nanovibrational bioreactor could be used to help provide humanised 3D models for drug screening. |

Results and Impact | ARCadia, was a festival of public events that celebrated the public opening of the ARC from 16-30 September 2022 that attracted almost 7000 people. The general aims were: Collaboration between the university and non-academic groups A mix of disciplines, from the arts to social, physical and life sciences Engaging traditionally marginalised groups, or groups who experience barriers to accessing the university Engaging communities local to the ARC (postcodes G11, G3, G12) 75 free events for all ages were held day and night in the ARC, from schools workshops to cinema screenings, exhibitions, comedy, gaming, talks and even an outdoor ceilidh with dancing dinosaurs. Over 63% of events involved working in partnership with non-academic groups. An astonishing number of people helped organise and run ARCadia events (and our group CeMi was central in this) , including 326 UofG colleagues, 147 students, 239 partner staff, 30 volunteers and for student interns. The event attracted public visitor of all ages throughout the 2 weeks and everyone provided good comments and they would like to be able to experience an event like that in the future. |

The aim was to raise as much as possible, a team from the CeMi run, walked and cycled the distance from Glasgow to the Sir Bobby Charlton Rehabilitation Centre in Amman, Jordan, that is 5436k! #RunToJordan We managed to raise £1,719! |

Results and Impact | Dr P.M. Tsimbouri has organised a full day workshop for P5,P6 and P7 school pupils at the Milngavie primary school. The workshop involves presentation slides and Q/A session with the children about the skeleton, bones and bone cells. This followed by hands on experience looking at fixed cells using bright field microscopes. The children were given colour-in pages and also word searches to take home. |
