Abstract

Technical Summary
The strategic objective of the LEGATO project is to promote the cultivation of grain legumes in Europe. Having identified the factors that currently limit grain legume cultivation, the project will devise solutions and assist the development of novel varieties and cultivation practices, targeted to food use. LEGATO will develop tools and resources to enable state of the art breeding methodology and to exploit fully the breadth of genetic resources available. The project will explore a number of key traits in studies that are complementary to other ongoing European and national projects, and include disease and pest resistance, where marker development for major fungal and viral pathogens plus a focus on emerging insect pests is planned. The impact of end-of-season drought and heat stress on the symbiosis with Rhizobium, and its consequences for plant performance, will be studied. Two major characters that influence grain legume yield, autofertility and number of flowering nodes, will be investigated. The potential for improving legume nutritional and organoleptic quality by identifying desirable traits and developing innovative selection methods will be investigated. LEGATO will develop and promote sustainable legume-based cropping systems adapted to different climatic regions, in considerations of local constraints. The project has been constructed around the participation of commercial partners, including SMEs, in the areas of marker development, plant breeding, and food processing, all of whom will benefit from advances in these areas delivered by LEGATO. Promising legume varieties and cropping systems will be tested at a series of pan-European sites in order to promote the widest possible uptake in agriculture. LEGATO partners will participate in a stakeholder forum which will convene regularly throughout the project.

Genetic diversity and strategies for seed quality enhancement in pea
| Description | Diverse germplasm was used to address questions surrounding the narrow range of genetic variation within breeding pools and to determine the distribution of allelic variants within such pools compared with wider germplasm. Novel variants for seed shape were identified and the genetic basis for the variation ascribed to a combination of loci. The data contributed to a publication (Rayner et al. 2017. International Journal of Molecular Sciences 18:1205. doi:10.3390/ijms18061205). The assays described in this publication for the r and rb loci in pea have relevance to many studies of the functional properties of pea products, specifically those carried out by LEGATO partners, with whom we are collaborating on a further publication. See Rayner et al. (2017) for a comprehensive analysis of the distribution of the naturally-occurring sbeI gene (r locus) among pea germplasm and its implications for the work of Mendel. In a collaborative study associated with the EU-funded project, the physicochemical functional properties of seeds from pea germplasm were examined (Santos et al., 2019). A collection of 105 pea accessions was analysed for physicochemical properties, pasting viscosity, and basic composition (protein, fibre, fat, and resistant starch). Phenotypic screens were revealed to be poor indicators of functional properties. Genetic screens, deployed on a subset of 32 lines, showed a correlation between genotype for sbeI and physicochemical properties, pasting viscosity, and basic compositional parameters (p < 0.05). |
| Exploitation Route | The information will be used by breeders and end users of legume products, in particular the food industry. Further collaboration with a group in Portugal has extended the value of the genetic assays developed to assessment of food quality traits (Santos et al., 2019). |
