Abstract

Technical Summary
The collaborative project between JIC and PGRO aims to provide growers with information about Downy Mildew (DM) race structure, geographical spread in the UK, varietal tolerance to races and the value of sources of resistance identified within the Pulse Crop Genetic Improvement Network across current UK growing areas. Pea DM is a major disease of both vining and combining peas in the UK and current conventional control options are limited to a seed treatment, rotational management and varietal tolerance. However there are reports that some varieties do not perform well in all pea growing areas due to the presence of different races of DM. The project aims to investigate pea DM population diversity in the UK and produce a map to illustrate relative occurrence of races and to support the later genetic studies. Current commercial varieties will be tested for susceptibility to different DM races to inform growers of varietal tolerance in specific areas. The information will be provided to growers to aid regional varietal selections. Improved knowledge of varietal selection will help to reduce the reliance on seed treatments.
In parallel, genetic studies will evaluate markers appropriate for the introgression of an identified resistance gene into commercial breeding stocks. Collaboration with the breeding industry will assist their selection and breeding programmes to develop improved resistance through genetic knowledge of current sources of resistance, alongside analysis of historical data relating to Pisum germplasm stocks, for which new molecular tools may now be exploited

Recombinant inbred lines derived from cultivars of pea for understanding the genetic basis of variation in breeders' traits
in Plant Genetic Resources: Characterization and Utilization
| Description | A source of downy mildew resistance in pea was identified, and genetic markers developed to assist breeders in using this and related lines in their breeding programmes. Genetic information and germplasm have been made available to breeders for use in programmes aimed at developing broad and durable resistance to downy mildew. The linkage group I genetic locus identified can be aligned with the syntenous region of the sequenced Medicago truncatula genome and this has permitted the development of gene-specific markers that work well for pea. These preliminary genetic data are being pursued within the research programme of the Defra-funded PCGIN, where it is clear that the same or a similar genetic locus exists in faba bean. The soil testing aspects of the work carried out by the PGRO partner within this project are relevant to pathology work being carried out within the Defra-funded PCGIN. The research alongside that of the Defra-funded PCGIN led to the formation of a consortium (JIC, NIAB, University of Worcester), which coordinated the submission of a LINK proposal to BBSRC on downy mildew in pea and faba bean, with considerable support from the industry. This project was selected for funding in 2020. Some of the marker data generated in the original project will be utilised by a new Group Leader at JIC whose focus will be on the genomics of disease resistance in pea. |

This project proposal includes developing some of the soil pathogen population aspects of the work, initially revealed by PGRO in the AHDB-funded research. The LINK project was approved for funding in 2020 and began in 2021. |

| Results and Impact | Meeting: Agri-Tech centre follow-up on sustainable pulse crop production and processing for the UK. First follow up call after group discussion on the 19th May. To further discuss and identify specific scope, missing partners and start thinking on activities/roles. UK's pulses production needs a downstream processing system including scale-up facilities for ingredient manufacturing and related research and development in ingredient functionality, with the focus on proteins. Crop varieties that are low/non-allergenic, have high yields, desirable functional properties and nutritious produce are needed to enable their upbuilt supply chain. Food manufacturers via consumers need to drive what direction the supply chain should focus their R&D efforts. End-product safety and quality will determine the required properties of ingredients, amd therefore the whole supply chain needs to work together as opposed to the current siloed approach. In order to develop a project, coverage from an end-user, food NPD, ingredient manufacturing (research and development, and scale-up), ingredient (protein) characterisation/optimisation (nutritional, functional), crop growing/producing and breeding perspective are needed. |
