Abstract

Technical Summary
The MLC is the hub of the new National Mouse Genetics Network - NMGN and therefore is tasked with ensuring cohesion and sharing of mouse resources, technical skills, scientific data and training throughout the whole network. Working alongside the director, Owen Sansom, we will ensure that strategies and programmes of work at the MLC follow the vision and emphasis of the Network leader.

| Description | Our previously published Mll-AF4 pre-leukaemia model was used as a platform in which to look for important co-drivers of leukaemia. We found that co-transfection of miR130b or miR128a produced a highly aggressive leukaemia phenotype with different lineage association. It also showed other important features of the disease such as CNS infiltration. This new model can be used for drug testing and to study the mechanism of leukaemia lineage choice and CNS disease. |

| Results and Impact | Rapid advances in gene editing and genetic technologies have revolutionised our ability to model human genetic disease and provided new hope for gene therapies. At this meeting, the very latest advances in modelling human genetic disease, from rare disease, cancer and immune disorders through to genetic modifiers were presented. They discussed the latest technologies and the challenges associated with generating relevant human genetic mutations in multi-modal systems, including advanced complex cellular models, model organisms and large animal models, and how genetic technologies can be applied to genome-wide screens and therapy. In addition, speakers outlined the latest strategies to model genetic mutations in cell type and tissue-specific mechanisms, in a developmental and time-dependent fashion, and in the coding and non-coding genome. Importantly, they discussed the latest approaches to the phenotyping of models and how to computationally align models with human disease to guide therapeutic intervention and, ultimately, translation. Event took place from 8 - 10 May 2024. Sara Wells was a panel member for this meeting. |
