Abstract
The Mary Lyon Centre at MRC Harwell (MLC) is the UK’s national facility for mouse genetics and the use of mouse models for the study of human disease. The specialised services available for UK researchers include the generation and study of genetically altered mice, and the National Mouse Archive, which allows scientists to deposit mouse strains free of charge for archiving and distribution purposes. The MLC is also the central hub of the MRC National Mouse Genetics Network which brings together experts from around the UK to collaborate and share resources.
Technical Summary
The Mary Lyon Centre (MLC) at MRC Harwell is the national facility for mouse genetics funded by the MRC. The MLC’s activities are focused on applying mouse genetics, transgenesis and phenotypic screening to investigate mammalian gene function at a systems level. We are working with genome editing technologies which are allowing us to introduce novel and/or complex genetic variants for preclinical and discovery research. Combined with our extensive expertise in cryobiology, rederivation and colony management for genetically altered (GA) strains, we are offering comprehensive science support for genetic research to both academia and industrial partners. The resources offered include:
• Generation of novel mouse models: Using current genome editing technologies
• Phenotyping and model characterisation: This includes comprehensive and in-depth platforms including imaging, metabolic and neurological phenotyping.
• Rederivation and cryopreservation of mouse colonies. The National Archive situated at the MLC allows the exchange of novel genetically altered stocks around the world, facilitating open science and the 3Rs.
• Quality Assured Animal Care: The Mary Lyon Centre is both ISO9001 and AAALAC accredited.
• Breeding and welfare assessment for GA colonies. Mouse strains are bred according to best practice, in order to maintain genetic integrity of mouse lines, prevent genetic drift or contamination and ensuring reproducibility.
• Therapeutic testing: Using newly developed disease-positioned, clinically focused mouse strains.
• In vitro platforms for cellular and expression studies
• Training: A innovative training centre (ADVANCE@MRC Harwell) opened in 2021, is building on the existing training offered in genetic science with an emphasis on in vivo research and technical training.
In addition, the Mary Lyon Centre is the central hub for the MRC’s National Network for Mouse Genetics (https://nmgn.mrc.ukri.org/). The Network consists of multiple disease-focused clusters pursuing distinctive, but complementary, challenge-led research and the Mary Lyon Centre at MRC Harwell acting as a central hub generating and robustly phenotyping strains, performing pre-clinical trials, and providing training. Key within these clusters are both experts in developing mouse genetic models, and translational and clinical scientists to provide a clear path to clinical translation. Furthermore, the clusters interact with each other, basing much of their in vivo work at the MLC, to capitalise on the numerous opportunities there will be for cross-disease collaboration and resource sharing.

| PI Contribution | Preclinical in vitro Models for the Replacement, Reduction and Refinement of Mouse Models in Preclinical Studies - Horizon - RIA project with 17 international partners from Germany, Netherlands, Italy, France, Check Republic, Finland, Denmark |
| Collaborator Contribution | The use of mouse models in biomedical research has been pivotal in understanding human diseases due to their genetic and physiological similarities to humans. However, concerns on ethics and translatability have prompted a shift towards alternative methods such as complex in vitro models (CIV Ms). PRIM-TECH3R addresses this shift by leveraging advanced technologies to develop reliable mouse and human CIVMs for preclinical studies, aiming to reduce and refine the use of animal models. Through standardised protocols and robust validation procedures, the project will create state-of-the-art in vitro models from mouse disease models, complementing existing in vivo models. This initiative aligns with EU policy objectives and societal demands to address phasing out of animal testing in biomedical research. The project will establish a comprehensive repository of preclinical resources, ensure open access, and foster collaboration between research infrastructures and the biomedical industry. Training programmes will equip personnel with the necessary skills to provide CIVMs to the research community. By achieving these objectives, PRIM-TECH3R aims to advance the state-of-the-art in research infrastructures, facilitate industrial exploitation, and contribute to transformative changes in biomedical research practices. |

| Results and Impact | 11 delegates attended the Mouse Embryo and Sperm Cryopreservation course. Cryopreservation allows long-term storage of embryos and sperm, providing a convenient way to preserve, protect and transport mouse strains used in biomedical research. Archiving scientifically relevant mouse lines also adheres to good animal welfare practices by removing the need for continued animal breeding and is integral to any comprehensive colony management programme. Running bi-annually since 2004, this course provides practical experience in murine embryo and spermatozoa freezing techniques routinely used at MRC Harwell and a simple, robust in vitro fertilisation procedure. The course will also cover the basic laboratory skills required when working in an assisted reproductive biology environment such as pipetting, weighing, microscope use and embryo handling. |

| Results and Impact | 14 delegates attended the Mouse Genetics and Colony Management for Researchers course. This course explores more complex genetic topics, including multiple allele crosses, complex disease models and will inform breeding strategies and experimental design rather than give a complete guide to the molecular biology. It examines how different modifications lead to different breeding strategies, the importance of genetic consistency and integrity and different modes of inheritance. There are multiple workshops throughout the course and a focus on troubleshooting unexpected issues when looking after Genetically Altered (GA) mouse lines. |
