Common translational mechanism's in fibrosis

Our preliminary data suggest that epigenetic modifications in fibroblasts may drive aberrant immune mediated tissue fibrosis and offers a potential for therapeutic manipulation of Dupuytren's disease. Similar studies from the Newcastle fibrosis Research Group have documented the importance of epigenetic mechanisms in liver fibrosis and highlighted that modulating epigenetic factors can suppress fibrogenesis. GSK has an extensive portfolio of novel compounds and tool products that target a range of fibrotic processes (e.g. BET family bromodomain inhibitors, immune modulators). This proposal will go further to determine the specific immune-mediated components and in keeping with the principles of EMINENT utilising the GSK clinic ready portfolio of potential immune modulator assets this could allow rapid identification and testing of potential mechanisms/therapies in novel human experimental systems pertaining to tissue fibrosis - DD thereby becomes an exemplar of accessible proof-of-mechanism human fibrotic tissue for subsequent extension to other relevant disorders across broader clinical fibrosis domains with the potential to become a further indication.
