Engineering synthetic post-transcriptional regulatory circuits in S. cerevisae for co-ordinated gene expression

Abstract

This project will explore options to develop novel forms of posttranscriptional circuitry for use in synthetic biology. In order to do this, an array of components, including RNA-binding proteins, RNA binding target motifs, degrons, effectors of these components, and other entities, will be studied in different combinations and tested in a range of novel scenarios.
The various components will be characterised in detail and then their behaviour will be studied in the context of newly-constructed circuits. One of the contexts explored will be that of regulated expression of heterologous genes, for example genes involved in the biosynthesis of mycosporin-like amino acids (MAAs).
In addition, the project will examine the benefits of combining transcriptional and posttranscriptional control elements in synthetic circuitry.
