Dissecting the molecular mechanisms by which Bmp8b increases thermogenesis in brown adipose tissue.

Abstract
Brown adipose tissue (BAT) is a unique tissue in that whilst it resembles traditional white adipose tissue in some ways it exists to burn fat rather than store it. Evolutionarily, BAT emerged as a way of generating heat to defend body temperature in cold conditions and order to perform this role it developed a number of specialized characteristics. BAT is heavily under the control of the sympathetic nervous system (SNS), to the degree that every single brown adipocyte receives neuronal input. This is so that the tissue can respond quickly to environmental demand, increasing mitochondrial content, mobilizing stored fat and oxidising large amounts of it to generate heat. To meet the demand placed on it BAT can expand in mass incredibly quickly, doubling in size and generating new nerves and blood vessels to supply it with the nutrients and control it requires. How all of these processes are activated, regulated and maintained is not understood but the mechanisms may be relevant to the regeneration of a range of tissues, for example veins, nerves and even liver.
We have discovered a molecule that is secreted into and around active BAT, which believe plays a crucial role in a number of processes. Our preliminary evidence suggests that bone morphogenetic protein 8b (BMP8b) is able to increase BAT sensitivity to activation by the SNS whilst directly regulating the growth of neurons and vascular cells. In addition, we have reason to believe that BMP8b also acts at a central level, regulating pathways in the brain that respond to environmental and dietary changes in order to increase SNS activation of BAT at the periphery. Understanding how a secreted molecule can elicit such specific effects in diverse cell types is likely to uncover new molecular pathways that co-ordinate the remodeling of mature tissues and their metabolic activity. It is also likely to lead to the generation of new targets for drug development in a range of diseases, even those mediated by aberrant sympathetic nervous system activity.
The potential benefits that may come from understanding the functions of BMP8b are not just extremely interesting from a molecular biology perspective. Recent studies have shown unequivocally that adult humans possess substantial quantities of brown fat. It may be that endogenous mechanisms such as Bmp8b exist in humans for activating BAT and increasing energy expenditure. In the face of a widening obesity epidemic, the ability to redress the positive energy balance that causes the disease would be extremely desirable.

Technical Summary
Brown adipose tissue is unique in its high oxidative capacity, mitochondrial content, sympathetic innervation and finally its ability to undergo rapid hyperplasia. The mechanisms regulating all of these processes, in a manner which coordinates multiple cell types are largely unknown in the mature organism. Bone morphogenetic proteins (BMPs) have been characterised as developmental factors that control cell differentiation and tissue organisation in embryogenesis. Bmp8b has two important features, distinct from other BMPs acting in adipose tissue. First, it is highly cold-inducible and second, it is expressed principally in mature brown adipocytes. Our preliminary data indicate that BMP8b is induced by cold in both classical brown and white depots in proportion to their capacity to form brown adipocytes. Mice over-expressing Bmp8b have increased thermogenic genes in subcutaneous WAT and levels of Bmp8b in vivo correlate with levels of neurogenic and angiogenic markers. Also, neuronal cells cultured ex vivo display increased growth and survival when treated with Bmp8b, suggesting BMP8b drives at least 2 of the three major processes (adipogenesis, neurogenesis and angiogenesis) involved in brown adipose tissue remodeling in response to cold. Through these processes and potentially also via its central actions, Bmp8b is able to regulate global energy balance, whereby Bmp8b -/- mice are susceptible to obesity.

Planned Impact
Medicine and Pharmaceutical Industry: Agonists of adrenergic receptors have been used successfully in the past to increase energy expenditure and treat metabolic disorders, however a lack of specificity has resulted in detrimental cardiovascular effects and the ultimate withdrawal of these drugs. Bmp8b represents a molecule that appears to act downstream of adrenergic receptors, opening up a novel molecular target (once the mechanism is better defined) for modulating the sympathetic nervous system sensitivity of specific tissues. Obviously, the role of Bmp8b in energy balance shown in our previous data makes it an attractive mechanism by which to examine the activation of human BAT, for the treatment of obesity. We already have strong links with UK based pharma companies as outlined in our proposal and as such our work could culminate in the generation of products which were of economic benefit to the UK as well as a health benefit to UK citizens.
Farmers: The ability to manipulate endogenous pathways that regulate energy balance may enable farmers to improve efficiency in terms of meat/dairy production offset to feed costs. In addition, the ability to enhance thermogenesis may improve survival and welfare in species reared outdoors in cold climates. In an increasingly competitive and highly subsidised industry, such advances would provide a economic benefit to the UK economy.
Food industry: Understanding how nutritional signals are integrated at a central level with activation/suppression of thermogenesis in BAT is likely to suggest dietary approaches to maintaining elevated energy expenditure in BAT. This is likely to be attractive to the "functional food" producers such as those making existing products enriched in omega3 fatty acids. Such products might offer a low-cost way to improve health and well-being in the UK without investment from central government. At the same time the products may themselves represent an economic benefit to the UK
Public health education: Our research could provide scientific background for public education on environmental and dietary stimulation of BAT to improve resistance to diet induced obesity. This could represent a significant benefit to public health and wellbeing.
The timescale for impact on most of the above is realistically in the order of 5-8 years for maximal effectiveness. However in the meantime the project will provide technological advances in real-time on an annual basis and provide a post-doctoral researcher (AW) with the skill necessary to advance his career to the young investigator stage. These skill would also be transferable to the private sector/pharma industry.

Description | We have learned about the role of BMP8b in adipose tissue contributing to control the adipose tissue function and remodelling. In adipose tissue BMP8b has a dual role. On one hand BMP8b contributes to thermogenesis through the direct action in the adipocyte to increase the sensitivity to adrenergic stimulation. This is important because the thermogenic activity of the adipose tissue requires less activation of the sympathetic tone to exert this effect. As the sympathetic tone also result in deleterious effects at the level of blood pressure and heart rate, the sensitising effect of BMP8b results in less requirement of adrenergic stimulation and potentially improved or ameliorated side effects. The other key observation is that BMP8b also contributes to the chronic remodelation of the beige adipose tissue by promoting the increase in vascularisation and innervation required for adipose tissue activation. The importance of this observation also is that we show that the adipocyte ad the needs imposed on it is a key determinant signalling and titrating the response of innervation and vascularisation to the functional needs of the adipose tissue. Also this research showed the concept of dormant cells differentiated to be beige cell but remaining in a dormant state waiting for the right moment to be activated when exposed to either cold or high fat diet. This opens opportunities to understand concept of programming of beiging of adipose tissue and the possibility that this may have memory. We have a paper just accepted in Nature Metabolism highlighting the role of the inducibility of BMP8b in liver as a factor that physiologically contributes to the healing of the liver but that when continuously induced due to sustain damage contributes to sustained inflammation and exacerbated liver damage. This awards has been also contributing to our research identifying molecular regulators of brown fat ( e.g. Thyroid hormone, Oestrogen receptor), teh contribution of specific types of fatty acids of different chain lenght (link with ELovl6), or nutrients such as Polyphenols. Simimilarly we have defined molecualr pathways regulated by lipids contributing to mitochondria activty and thermogenesis. We also idetified inactivators of brown fat such as LR11, which are potentially important to prevent excessive energy dissipation contributing patholies such as anorexia or cancer cachexia treatment. I think this award was important for my laboratory to obtain a position of leadership in the field at international level. Also has contributed to create opinion through reviews and conferences. Finally I think the benefit to the development of the PhD students and postdocs has been important.. |
Exploitation Route | This information is essential to consider BAT a potential therapy for obesity in the context of expanding or activating BAT. Also the knowledge generated has helped us to secure funding through ERC senbior investigator to focus on the role of human Pluripotent stem cells to differentiate into human brown fat cells and also to obtan funding from IMI Litmus project to continue work on fatty liver. Also the knowledge obtained suggests that BMP8b may also contribute to apsects of liver cancer. This is an area that will be followed by one of the Post docs when applying for his career development fellowship. Results and Impact | New protein could help burn fat faster By Meera Senthilingam, for CNN Updated 1109 GMT (1909 HKT) February 1, 2016 (CNN)-Research has shown that the fatter your body becomes, the harder it becomes to burn that excess fat. As people eat more fat than they burn, the body stores this excess in various parts of the body and as this storage increases, metabolism changes. However, all is not lost: a recent study by researchers at the University of Cambridge identified one of the proteins underlying this blockage of fat burn. "[This protein] is part of the system in place to help the body store more energy," says Antonio Vidal-Puig, Professor in Molecular Nutrition and Metabolism, who worked on the study. The culprit, called sLR11, was found to block the functioning of brown fat cells -- responsible for generating heat to keep us warm -- and instead help the body store fat more efficiently by preventing excessive heat generation. The benefits of brown fat There are two forms of fat cells -- brown cells and white cells -- and each plays a different role in our metabolism. While brown cells help generate heat, the white cells are responsible for the storage of fat -- or energy -- ready for release as needed. Levels of brown fat are known to be high in children but recent findings showing the presence of brown fat in adults, restored hope to use them as targets to treat obesity. According to Vidal-Puig, obese people have less brown fat than lean people -- and the same applies for diabetics. "If you can increase the brown fat in obese and diabetic people then you can improve their situation," says Vidal-Puig. This has been known for some time, but making it a reality has been a challenge. The newly discovered sLR11 protein could provide the answer. Removing the brakes Vidal-Puig compares the protein to the brake of a car, stopping the body from accelerating its production of heat. "Obese people have more of these brakes, preventing them from creating heat and therefore losing fat," says Vidal-Puig who has seen elevated levels of the protein in the blood of obese people included in his study. During the study, the team removed the gene producing this protein in mice and found they were able to burn calories faster. The researchers went on to examine the protein in humans and found that levels of the protein correlated with the total fat mass in a person. What's more, investigated levels in obese patients after they underwent bariatric surgery, and their weight loss was directly proportional to the decrease in levels of the sLR11 protein. "[Brown fat cells] are an attractive therapeutic target for weight loss, and for anti-obesity or anti-type 2 Diabetes (T2D) therapies," says Carole Sztalryd, Associate Professor of Medicine at the University of Maryland School of Medicine. Sztalryd's team also work on biological approaches to weight loss. More brown than white on the cards A common focus to tackle obesity has been on accelerating the production of brown fat cells so people burn more calories, but this new insight could achieve the same but through enhancing the activity -- and efficiency -- of cells. "We are stopping the brakes within the fat cells," says Vidal-Puig, as the mice in his study became protected from obesity with increased browning of their white fat cells. The team believe controlling energy storage will be a better approach to beat the obesity epidemic than controlling people's eating habits. But Sztalryd warns that additional studies are required to evaluate any clinical relevance,"New therapies are urgently needed for obesity. sLR11 seems promising but more work is needed to confirm if this is a relevant," she says. In the mean time, old tactics may be the answer and the more weight gained, the more exercise to be done. |

Results and Impact | EAS Advanced in Cardiometabolic Phenotyping in Rodents, Cambridge, UK Scientists from the University of Cambridge Biomedical Campus recently hosted the 2nd EAS Advanced Course in Cardiometabolic Phenotyping in Rodents. This 3-day course combines the state of the art facilities and expertise within the Disease Model Core at the WellcomeTrust-MRC Institute of Metabolic Science and the BHF Cardiovascular Centre. By integrating the expertise of these two specialized phenotyping centres, it provides a unique opportunity to learn about many of the skills required for in depth cardiometabolic phenotyping in small animals. The 32 participants, selected from centres across Europe, were treated to a programme of lectures, phenotyping demonstrations and data analysis sessions. A further 1-day workshop at the Sanger/EBI focused on mouse genetics, genetic engineering and informatics resources, highlighting the great opportunities provided by recent advances in genetic engineering purpose-made mouse models. Participants particularly appreciated the mixture of hands-on experience, combined with opportunities to ask detailed questions on how the techniques demonstrated should be used to suit their own research priorities. The course was sponsored by the European Atherosclerosis Society, British Heart Foundation and TSE systems, with organisation led by Prof Toni Vidal-Puig, Director of the MRC Metabolic Diseases Unit Disease Model Core. |

La grasa parda es un órgano diseñado para producir calor. Está presente en los recién nacidos porque son muy vulnerables frente al frío, y tienen que producir calor para mantener la temperatura corporal adecuada. Lo que se ha observado recientemente es que también la tienen algunos adultos. El equipo del profesor Vidal-Puig y otros están estudiando la posibilidad de desarrollar un fármaco capaz de estimular la producción de grasa parda. Results and Impact | On Monday February 17th 2020, a team of PhD students from the Institute of Metabolic Science went to the Suffolk Science Festival in Bury St. Edmunds to teach local children about neurons, the anatomy of the gastrointestinal system and the importance of insulin sensitivity. Among the team was TVP lab PhD student Jennie Leggat. |

Spiegelman and Sven Enerbäck April 17-22, 2015 Snowbird Resort • Snowbird, Utah USA The epidemic of obesity and diabetes has become a world-wide health risk. These metabolic disorders are a direct threat to the health and economic well-being of many nations. Unfortunately, education about diet and exercise are not sufficient to meet this emergency and new therapies are badly needed. Brown fat has emerged as one of the few genuinely new approaches to these problems. Brown fat exists in most or all adult humans and can dissipate caloric energy and improve both obesity and diabetes in experimental animals. This meeting will focus on new pathways controlling brown fat development and thermogenesis. We will delve into the types of brown fat cells, beige, classical brown and others. We will also put a special emphasis on emerging approaches, both biological and chemical, that can manipulate these cells for therapeutic purposes. It is our hope and belief that this meeting can nucleate expanded efforts in both academia and pharma to explore this promising research area. There has not been a Keystone Symposia conference dedicated to brown fat. While there has been a session here and there at the Obesity meeting, the science and the biomedical needs warrant this new and focused conference. Of course, this will also bridge the Obesity and Diabetes communities, and lead to pharma/academic collaborations. |

In spite of all the millions spent on diets, food and exercise, it may be that something as basic as temperature is the answer. We all have two types of fat, white and brown. White fat is inert and sits in deposits. Brown fat, which regulates babies' temperatures but is much harder to pinpoint in adults, can be activated to burn calories and generate heat. Studies show that exposure to low temperatures may encourage white fat to behave like brown fat. So while central heating ensures we move less and burn fewer calories, it's possible that lower temperatures might increase the activity of brown fat, which will burn our calories for us. |
