Using mathematical modelling of the Langerhans cell tolerogenic gene regulatory network to identify key molecular regulators of the tolerogenic progra

Lead Research Organisation: University of Southampton
Department Name: Clinical and Experimental Sciences

Abstract

Skills Priority Alignment: Quantitative Biology

Hypothesis
LC tolerogenic ability is regulated by a unique gene regulatory network (GRN), which induces a distinct gene expression profile associated with the tolerogenic LC programme.

Aims
1) Investigate cell-cell variation within LC populations.
2) Dissect a unique gene expression profile associated with tolerance in LCs and identify the TFs essential for regulating LC tolerogenic function.
3) Create a mathematical model of the LC tolerogenic GRN for quantitative in silico simulations that correctly predict the expression of tolerogenic output genes
4) Identify the key molecular switches regulating tolerogenic responses and validate in vitro.
