Solid polypeptide dosage forms for transdermal and subcutaneous administration

Abstract

The project aims to develop polypeptide (peptide and protein) formulations for transdermal and subcutaneous (sc) delivery. Model polypeptides will be formulated and processed with excipients that have been shown to stabilise proteins as solid particles. Drug loaded particles will be fabricated using emulsion and electrospraying processes. The excipients will include common polymers and low molecular weight molecules (e.g. buffers, amino acids). We will explore formulation and processing strategies to achieve controlled and extended release while maintaining polypeptide function. Dosage form design for administration of the solid, stabilised particles will be examined including microneedles for transdermal administration and as an subcutaneously injectable particle depot system.
