Abstract
This project involves using proteomic and metabolomic approaches to analyse post-mortem human brain tissue to determine molecular changes that occur in various neuropathological disorders. In partnership with Bruker Daltonics, we will aim to develop workflows that integrate Mass Spectrometry Imaging using the next-generation MALDI-2 instrument with other tissue analysis methods, such as histology, laser capture microdissection, and LC-MS/MS. In doing so, the aim is to resolve tissue- and disease-specific molecular pathways in a spatial context, and to develop high-throughput methods that can be used for biomarker validation. Moreover, the project will contribute to the improvement of the analytical challenges that these next-generation instruments present, including sample preparation, high-throughput, data acquisition, and interpretation, ultimately developing the prospect of these instruments to revolutionise brain tissue diagnostics in the clinic.
