Coordination of localized actin polymerization with endosomal membrane dynamics

Abstract

For cells to respond to their environment, they must employ several pathways to move molecules to the correct destination within the cell where signals can be processed, and their behaviour can be adjusted in response. Once molecules are internalized, they are sorted into different pathways from sorting compartments. However, how the cell is capable of recognizing and sorting molecules into the correct pathways remains unclear. Recent evidence implicates the WASH generated actin network in the function of these sorting compartments. Using a cryogenic correlative imaging approach, we will be able to observe this system with a novel perspective to gain insight into the involvement of WASH and its actin network in this sorting process. This will improve our understanding of cell signalling and how cells adapt in response to environmental cues, which will be essential as errors in signalling processes are responsible for numerous pathological conditions. Additionally, our work includes a component of development of optimized cryogenic correlative imaging equipment and procedures for the evaluation of emerging imaging technology.
