Determining the relationship between TFEB and the NSL complex in relation to the regulation of mitophagy

Lead Research Organisation: UNIVERSITY COLLEGE LONDON
Department Name: Cell and Developmental Biology

Abstract

Our previous work identified KANSL1 and KAT8, two components of the NSL chromatinremodelling
complex, as transcriptional regulators of mitophagy. Our unpublished
RNA-sequencing data further reveal an enrichment of genes regulated by TFEB, a
master regulator of lysosomal biogenesis / autophagy[1], in KANSL1 knockdown (KD)
cells. Finally, interrogation of publicly available TFEB transcription regulation
databases suggests that three regulators of the NSL complex are themselves
regulated by TFEB. Taken together, these data suggest a functional relationship
between TFEB and the NSL complex, which could inform a more complete
understanding of the regulatory network governing the balance between, mitophagy,
autophagy and lysosome biology.TBC
