Soft-Hard Tissue Engineering for Synthetic Bone Manufacturing using Ultrafast Lasers

Abstract
There is a huge shortage of bio-compatible bone materials in the world for replacing damaged bone tissue. The PhD topic will focus on a novel method for synthetic bone materials engineering using ultrafast lasers and advanced bio-materials. The multidisciplinary research will focus on the sequential deposition of calcium phosphate minerals with collagen, and protein and growth factor gene. The method of bone materials fabrication by engineering hard (mineral)-soft (collagen, protein, growth factor) will also examine micro-mechanics, structure, and bio-compatibility for synthetic bone manufacturing. The manufacturing engineering will also be supplemented by training in cell viability tests. In the project the student will be trained in the core areas of research in laser processing, characterisation, cell viability and micro-mechanics. The PhD student will also gain some experience by working with clinicians.

Description | The EPSRC project is focused to establish a novel approach, for the development, fabrication and implantation of bone scaffolds, based on the fundamentals of advanced manufacturing using femtosecond lasers. Bone is a complex living tissue with significant metabolic and regenerative activities, which are disrupted when the tissue is damaged. Healing of fractured bone is a complicated process therefore; an ideal bone scaffold should have: i) exceptional osteogenic potential in order to promote new bone formation; ii) load-bearing properties; iii) appropriate microstructure for promoting angiogenesis for the circulation of nutrients and iv) antibacterial resistance to avoid infections that lead to failure of the surgery. Research efforts have been focused on development of the "perfect" biomaterial, targeting to achieve all the aforementioned properties with a single material system; e.g. blocks of calcium phosphates, coated Ti-implants, polymers etc. Micropatterning and microchannels promote guided cellular growth leading eventually to the formation of vasculature. In addition, the increased capillary forces in the channels allows for better circulation of the nutritional components which are necessary for the formation of new blood vessels. |
Exploitation Route | Further investigation into nanoparticles which are emerging as novel antibacterial agents as they have proven their effectiveness for treating infectious diseases. Factors such as size, shape, surface charge and surface area to volume ratio affect the antibacterial potential of nanoparticles although the exact antibacterial mechanisms are not yet fully clear. |
