Abstract
The proposed Platform grant renewal is aimed at giving us the ability to capitalise upon our position as a world leading group in membrane biophysics and self-assembled amphiphilic materials, and to promote and facilitate collaborations within the currently fragmented membrane biophysics community in the UK. To achieve these aims we will form collaborative research networks with groups having complementary expertise, and encourage interconnections between external groups via a combination of 'Ideas Workshops' and IT platforms. These networks will focus upon tackling key challenges at the Life Sciences Interface requiring multi-disciplinary approaches. We will support targeted short-term personnel exchange with a view to effecting knowledge transfer, proof-of-concept experiments and research training. Within this framework we will invite key collaborators as academic visitors and to present seminar series within our Department. We also wish to continue to facilitate exploitation of our specialised facilities by the wider research community.In addition to building upon the successes of our original objectives by studying the phase behaviour of novel amphiphiles and complex lipid mixtures, and the development of millisecond and microsceond pressure-jump synchrotron X-ray studies of lyotropic transitions, we also plan to develop the following areas of research within the renewed Platform grant, which we have identified as timely and important challenges at the Life/Physical Sciences interface:i) Understanding Membrane Asymmetry;ii) Protein Control of Lipid Membrane Morphology;iv) Regulation of Lipid Biosynthetic Pathways.iv) Lipid Mesophase Engineering Rules for Biomedical Imaging Applications.

Description | A selection of Key Findings from our Platform grant is as follows: a) We have demonstrated that lipid membranes can act as information pathways between proteins, without the need for direct protein-protein contact; b) We have found that lipid membrane curvature is induced by distearoyl phosphatidylinositol 4-phosphate; c) We have characterised the pressure - temperature phase behaviour of natural sphingomyelin extracts, and found evidence for non-lamellar phases; d) We have characterised the thermotropic and lyotropic liquid crystalline phase behaviour of a novel series of Guerbet branched-chain ß-D-glucosides, and have found that they form a variety of inverse cubic and inverse hexagonal phases; e) We discovered a novel lyotropic two-dimensional inverse ribbon phase in a branched-chain polyoxyethylene surfactant system, and demonstrated an unusual effect of hydrostatic pressure on the lattice parameters, with one increasing, and the other decreasing, with increasing pressure; f) We showed that we can produce ordered nanostructured amphiphile self-assembly materials from nonionic unsaturated monoethanolamide lipids in water; g) We studied the effect of hydrostatic pressure effects on the inverse micellar Fd3m cubic phase formed by a binary lipid mixture, and found that the lattice parameter is independent of pressure, but above a threshold pressure the lipids separate into two different liquid crystal phases; h) We developed a quantitative model for the kinetics of lyotropic phase transitions involving changes in monolayer curvature; i) We showed that certain aqueous buffers may adversely affect the phase behaviour of model lipid membranes; j) We have a developed a new model for how cationic drugs can interact with lipid membranes; k) We carried out molecular dynamics simulations of liquid condensed to liquid expanded transitions in DPPC monolayers; l) We observed the formation of an inverse hexagonal phase in DNA Lipoplexes by molecular dynamics simulations; m) We developed a microfluidic approach for high-throughput droplet interface bilayer (DIB) formation; n) We developed a microfluidic antibody capture chip with TIRF detection as a first step towards practical single cell proteomics. |

Symposia were showcasing various aspects of collaboration across different geographic regions or between different sectors of chemical research (industry, academia, research institutes, government laboratories). Symposia were focusing on partnerships between traditional industries and contract research organizations (CROs) or contract manufacturing organizations (CMO's). Other collaborative arrangements of interest included precompetitive industrial collaborations on enabling chemical technologies, new technologies and new strategies for facilitating effective collaborations across multiple time zones, and managing intellectual property and licensing. |

Results and Impact | Organised the London Schools Chemistry Challenge, a competition designed to commemorate the International Year of Chemistry that was open to all students studying Chemistry (GCSE or A-level) in secondary schools in Greater London. Entries could take the form of a poster, model, booklet, video presentation-the choice was left to the students. The competition was an overwhelming success with 1300 students registering to take part from 250 schools across London. |
