Abstract

The Enabling Technologies programme consists of a number of core facilities that provide critical technologies and equipment to scientists in the MDU and it is crucial its success. Because metabolic disease affects the entire body, it is vital to study these diseases whole animals. Our model of choice is the mouse. The Disease Model Core provides facilities for the measurement of the amount of fat and muscle in an animal’s body, how much and what type of food is eaten, and how much energy is burnt, through either metabolism, heat production, or exercise. It also provides facilities for examining isolated cells, tissues and organs. At the other end of the size spectrum, the Genomics/Transcriptomics Core enables scientists to study the sub-microscopic world of genes and how they function and are turned into proteins and the Core Biochemistry Assay Lab measures hormones and other molecules in blood.

Technical Summary

This programme provides the core facilities and technologies that act as vital supporting infrastructure, not only for MDU Programmes 1-4 but also for the other investigators across the MRL. At the heart of this programme are the Disease Model Core (DMC), the Genomics and Transcriptomics Core (GTC) and the Biochemistry Assay Lab (CBAL).

The DMC integrates two major platforms, physiology and histology, and provides a full range of support services to maximise scientific output from in vivo model systems. These include Home Office licensing support, cutting-edge in vivo phenotyping techniques, ex vivo tissue analysis, statistical analysis and data interpretation. The GTC provides access to high-throughput sequencing technologies, RNA quantification and ‘Sanger’ DNA sequencing as well as bioinformatics support. CBAL runs alongside the Clinical Biochemistry Lab of the Cambridge NIHR Cambridge Biomedical Research Centre and offers biochemical and immunoassay analyses (mainly rodent plasma and serum) to members of the MDU and IMS-MRL.

While the DMC and GTC are the primary facilities within Programme 5, a number of other key technologies, which receive the majority of their funding from other sources (Wellcome Trust and NIHR BRC), also provide critical support to MDU scientists. These are Microscopy and Imaging, Metabolomics/Lipidomics, Proteomics and the Genome Engineering Core. We seek to build on the success of these cores facilities and strengthen key areas where there is pressing, multi-user need.

Title FM, quantitative mass spectoscopy for analysing neuropeptide production 
Description We used liquid chromatography tandem mass spectroscopy (LC-MS/MS) to develop quantitative methods to analyse neuropeptide production in cultured human cells and in primary human brain samples. This consists of both standard curved made with sythetic human peptides, and synthetic peptides generated to incorporate stable heavy isotopes, which are spiked into samples during preparation. This enables very accurate and sensitive quantification of peptides, and assessment of relative quantities of different peptides of interests.

Impact This method enabled us to examine the relative production of POMC-derived peptides in both human cellular culture and in the primary brain. We found that alpha-MSH, which is the predominant MSH form studied, is produced at most lower levels than desacetyl alpha-MSH. Furthermore, we confirmed that beta-MSH, whose existence has been controversial, is indeed produced both in vivo and in vitro at similar quantities as desacetyl alpha-MSH.

Title High-throughput single cell unbiased lipidomics assay in human iPSC-derived dopamine neurons (AK) 
Description The development of single cell lipidomics approaches to study the lipid metabolism of dopaminergic neurons in relation to parkinson's disease. Changes in lipid metabolism have been strongly associated with Parkinson's disease (PD). Lipids have the potential to be important biomarkers for PD, but the lack of appropriate tools to measure total lipids (lipidome) in human dopamine neurons has limited advances in this field. we have developed a novel unbiased and high-throughput single cell assay to determine total lipids. This will allow us to identify and validate lipid metabolites in dopamine neurons, which may serve as biomarkers for PD.

Title bioinformatic pipeline for analysis of sequencing data from gene-edited cell lines (FM)

To address this issue we developed GenEditID, a flexible, open-access platform for sample tracking, analysis and integration of multiplexed deep sequencing and proteomic data, and intuitive plate-based data visualisation to facilitate gene edited clone identification.

Title Alteration of the plasma peptidome of gastrectomised patients during an OGTT 
Description Performing gastric bypass surgery on patients severely changes their gut hormone response to glucose. The plasma concentrations of Insulin and a number of associated incretin peptides are significantly raised shortly after ingestion of glucose. Typically the plasma concentrations of these peptides are monitored using immunoassays. A simple and high throughput plasma peptidomics approach was used to simulatneously monitor a large number of these peptides. Comparison of gastresctomised and control individuals showed significant increases of insulin, glucagon gene products, PYY and neurotensin.

Title Expected values for gastrointestinal and pancreatic hormone concentrations in healthy volunteers in the fasting and postprandial state 
Description Background: Gastrointestinal hormones regulate intestinal transit, control digestion, influence appetite and promote satiety. Altered production or action of gut hormones, including glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP) and peptide YY (PYY), may contribute to the biological basis of obesity and altered glucose homeostasis. However, challenges in analytical methodology and lack of clarity on expected values for healthy individuals has limited progress in this field. The aim of this study was to describe expected concentrations of gastrointestinal and pancreatic hormones in healthy volunteers following a standardised meal test (SMT) or 75g oral glucose tolerance test (OGTT). Methods: 28 healthy volunteers (12 men, 16 women; mean age 31.3 years; mean body mass index 24.9 kg/m2) were recruited to attend a hospital clinic on two occasions. Volunteers had blood sampling in the fasting state and were given, in randomised order, an oral glucose tolerance test (OGTT) and standardised mixed liquid meal test with venepuncture at timed intervals for 4 hours after ingestion. Analytical methods for gut and pancreatic hormones were assessed and optimised. Concentrations of gut and pancreatic hormones were measured and used to compile ranges of expected values. Results: Ranges of expected values were created for glucose, insulin, glucagon, GLP-1, GIP, PYY and free fatty acids in response to a standardised mixed liquid meal or OGTT. Intact Proinsulin and C-peptide levels were also measured following the OGTT. Conclusions: These ranges of expected values can now be used to compare gut hormone concentrations between healthy individuals and patient groups.

Title Glucose-dependent Insulinotropic polypeptide receptor-expressing cells in the hypothalamus regulate food intake 
Description Ambiguity regarding the role of glucose-dependent insulinotropic polypeptide (GIP) in obesity arises from conflicting reports asserting that both GIP receptor (GIPR) agonism and antagonism are effective strategies for inhibiting weight gain. To enable identification and manipulation of Gipr-expressing (Gipr) cells we created GIPR-Cre knock-in mice. As GIPR-agonists have recently been reported to suppress food intake we aimed to identify central mediators of this effect. Gipr cells were identified in the arcuate, dorsomedial, and paraventricular nuclei of the hypothalamus, as confirmed by RNAscope in mouse and human. Single cell RNAseq identified clusters of hypothalamic Gipr cells exhibiting transcriptomic signatures for mural, glial and neuronal cells, the latter expressing somatostatin, but little proopiomelanocortin or agouti-related peptide. Activation of Gq-DREADDs in hypothalamic Gipr cells suppressed food intake in vivo, which was not obviously additive with concomitant GLP1R activation. These data identify hypothalamic GIPR as a target for the regulation of energy balance.

Title Peptidomic analysis of pancreatic neurodendocrine tumours (Glucagonoma) 
Description A peptidomic analysis of plasma from patients with well characterised pancreatic neuroendocrine tumours (PNET) was performed. A nano LC-MS analysis identified a number of peptides from the glucagon gene, which were idenfiried in a previous case study using multiple immunoassays. Peptides to other proteins involved in peptide secretion were also identified. Plasma from a second glucagonoma patient was analysed using a high flow approach, and this identified similar peptides to the nano analysis. In order to demonstrate the potential clinical application of LC-MS to characterising neuroendocrine tumours, a large cohort of plasma samples were analysed to demonstrate the ability of this approach to identify glucagonoma patients, and differentiate from a single insulinoma patient

During the lockdown, two members of the TVP Lab - Sonia & Jennie - took part in the MRC initiative 'I'm a Scientist - Stay at Home!'. This programme was established to allow school children to talk to real scientists during their virtual science lessons in the summer term, to find out more about the subjects they were interested in! Sonia was in the 'Disease Detectives Zone', while Jennie was in the 'Medical Research Zone'. They both had a lot of fun, and were amazed by how insightful the questions they received were!

However, this turbulent time has also enabled new, unexpected leaders to emerge, performing more autonomously and making insightful decisions. Results and Impact IMS staff and students worked with a local secondary school teacher and a communications trainer to develop 1 existing and 2 new activities for use with Year 9 pupils. We ran a whole day of activities on a bus with each of 2 schools considered to have 'under-served' intake - North Cambridge Academy and Ely College - meeting the complete Year groups in each case. We reached over 300 teenagers with the activities.

Feedback from our researcher volunteers:
- [Teachers] were also very complimentary and engaged with our activity as it touched on their lessons or personal lives
- I was surprised at just how smoothly it went, and by how engaged the pupils were Several teachers commented that they really liked the fact that we had three different activities that looked at different areas of research, but also themselves appealed to different personalities
- I was surprised how children very suddenly came up with very interesting and smart questionsit definitely makes you think about the scientific content in a different way!
- I learnt that secondary school children are being taught more complex science than I expected

Post-event responses from 62 year 9 pupils at North Cambridge Academy showed that
- 87% learned something new
- 79% did not have any relative or friend who worked in science or for a university
- just 11% (7 students) remembered ever going to the Cambridge Science Festival.
- 25% said that after the session they were more likely to consider a career in science.

Feedback on leaving the Institute from a staff member involved in development and delivery of this activity:
- Taking part in PE helped me to decide about the job change. I got the new job partly because of the skills and experience from being involved

Results and Impact Request was: I am currently researching for a TV report about the links between obesity and COVID-19. This is prompted by a recent study from Italy in the European Society of Endocrinology, suggesting that people with a BMI of over 30 are at a greater risk of severe COVID-19 infection. There seems to be a renewed discussion for a "weight-loss drive" in anticipation of second wave of coronavirus later in the year, and we are keen to delve further into this. The report itself will be for our U.S. output. I was wondering if you might be available to speak to us about your understanding of the links between obesity and COVID-19, initially for a research call, but with the view of arranging a video interview? We are keen to learn more from your expertise on this subject.

Results and Impact VBL presented a poster at a local Cambridge Neuroscience Association meeting at Robinson College on March 14th, 2019. It was called "Cambridge Neuroscience Seminar 2019: Signalling, Sensation & Sentience" and the poster title was "ATP is released from GLP-1 secreting enteroendocrine cells and signals to vagal afferent neruones". It was a valuable experience sharing our work on gut-brain signalling to the larger Cambridge neuroscience community.
