Photo-Electro: Transforming Chemical Synthesis, Discovery and Manufacture
Lead Research Organisation:

Department Name: Sch of Chemistry
Abstract
Our vision is to use continuous photochemistry and electrochemistry to transform how fine chemicals, agrochemicals and pharmaceuticals are manufactured in the UK. We aim to minimize the amount of chemicals, solvents and processing steps needed to construct complex molecules. We will achieve this by exploiting light and/or electricity to promote more specific chemical transformations and cleaner processes. By linking continuous photochemistry and electro-chemistry with thermal flow chemistry and environmentally acceptable solvents, we will create a toolkit with the power to transform all aspects of chemical synthesis from initial discovery through to chemical manufacturing of high-value molecules.
The objective is to increase efficiency in terms of both atoms and energy, resulting in lower cost, low waste, low solvent footprints and shorter manufacturing routes. Historically photo- and electro-chemistry have been under-utilised in academia and industry because they are perceived to be complicated to use, difficult to scale up and engineer into viable processes despite their obvious environmental, energy and cost benefits. We will combine the strategies and the skills needed to overcome these barriers and will open up new areas of science, and deliver a step-change (i) providing routes to novel molecular architectures, hard to reach or even inaccessible by conventional methodologies, (ii) eliminating many toxic reagents by rendering them unnecessary, (iii) minimizing solvent usage, (iv) promoting new methodologies for synthetic route planning. Our proposal is supported by 21 industrial partners covering a broad range of sectors of the chemistry-using industries who are offering £1.23M in-kind support.
Therefore, we will study a broad range of reactions to provide a clear understanding of the most effective areas for applying our techniques; we will evaluate strategies for altering the underlying photophysics and kinetics so as to accelerate the efficiency of promising reactions; we will transform our current designs of photochemical and electrochemical reactors, with a combination of engineering, modelling and new fabrication techniques to maximize their efficiency and to provide clear opportunities for scale-up; we will exploit on-line analytics to accelerate the optimisation of continuous photochemical and electrochemical reactions; we will design and build a new generation of reactors for new applications; we will identify the most effective strategies for linking our reactors into integrated multi-step continuous processes with minimized waste; we will demonstrate this integration on at least one synthesis of a representative pharmaceutical target molecule on a larger scale; we will apply a robust series of sustainability metrics to benchmark our approaches against current manufacturing.

Planned Impact
Our Programme, Photo-Electro, will have substantial impact on chemists in academia and industry because we will demonstrate the value of continuous photo-, electro- and thermochemistry to make complex molecules. Making such molecules is a key activity in a wide range of UK chemistry-using industries. These include pharmaceutical and agrochemical manufacture as well as companies making speciality chemicals for fragrances and many other end products. Such industries are under constant regulatory and financial pressure to develop processes that are environmentally and economically more efficient than current methodologies. Therefore, our approach of replacing toxic and/or wasteful stoichiometric reagents by light, electricity and heat, has great attractions as demonstrated by the letters of support from our 21 industrial partners.
What makes our programme different is that we are using light, electricity and heat together to deliver integrated processes to carry out multi-step assembly of complex molecules. At the same time, we are designing and building new continuous reactors which, by the end of the Programme, will be available to non-specialist chemists in industry and academia. The designs will be informed by computer fluid modelling and chemical engineering so that we maximise the efficiencies of both mass and heat transfer. Our reactors are intended to be both reliable and scalable so as to simplify their eventual introduction into commercial chemical production. We have a robust strategy for impact. Our industrial partners will form a stakeholders' forum to engage with our research right from the start. The Forum is intended to be inclusive and new members will be warmly welcomed at any stage of our programme. Photo-Electro is particularly timely as it will deliver new methodologies which can increase the competivity of UK chemical manufacturing. And, in this period of post-referendum uncertainty, increasing that competivity must be a national priority.

Using batch reactor results to calculate optimal flow rates for the scale-up of UV photochemical reactions
in Reaction Chemistry & Engineering
Description | • New designs of reactors, particularly a family of Taylor Vortex Reactors, optimised using Computational Fluid Dynamics. They suitable for both UV and visible photochemistry up to scales equivalent to 11 kg/day and for electrochemistry up to 1 kg/day. • Telescoped photochemical and thermal reactions, where solvents superheated above their normal boiling points in pressurised tubular reactors have accelerated thermal reactions from times of hours to only minutes allowing the reactions to carried out efficiently as a flow process. Thereby allowing previously unproductive reactions to reach synthetically useful yields. • Application of time-resolved infrared and UV-visible spectroscopies for understanding the kinetics of complex photochemical reactions. • Application of process analytical technologies including FTIR, Raman and GC for optimisation of flow chemistry under AI control. • New application of A-TEEMS absorption/emission spectroscopy for monitoring continuous organic chemistry with high sensitivity. • New or greatly improved photochemical reactions, including the use of thiaxanthone photosensitisers which switch reactions from the UV to visible regions. • New or unexpected photochemical reactions carried out in flow for the first time. • New continuous electrosynthetic reactions which do not require the use of divided electrochemical cells. |

Description | Photo-Electro was a Programme Grant involving chemists and engineers at three universities, Bristol, Nottingham and Southampton, seeking to transform the manufacturing of fine chemicals, pharmaceuticals and agrochemicals by using photochemistry and electrochemistry. Our aim has been to reduce the use of solvents, chemicals and processing steps needed to synthesize complex molecular structures. The objective is to increase efficiency in terms of both atoms and energy, resulting in lower cost, low waste, low solvent footprints and shorter manufacturing routes. Historically photo- and electro- chemistry have been under-utilised in academia and industry because they are perceived to be complicated to use, difficult to scale up and engineer into viable processes despite their obvious environmental, energy and cost benefits. We have combined the strategies and the skills needed to overcome these barriers and will open up new areas of science and deliver a step-change (i) providing routes to novel molecular architectures, hard to reach or even inaccessible by conventional methodologies, (ii) eliminate many toxic reagents by rendering them unnecessary, (iii) minimizing solvent usage, (iv) promoting new methodologies for synthetic route planning. Our project has been very timely because it has coincided with a major resurgence of interest in both photochemistry as a synthetic tool and in electrosynthesis. Our new reactors, particularly the so-called Taylor Vortex Reactor, have attracted widespread interest from academia and industry. The novelty of our Vortex design is that it can be applied to photo-, electro- and thermos-chemical reactions in flow and, unlike most continuous reactors, residence time and mixing are largely decoupled in the Vortex Reactor. The design won first prize in the 2023 Royal Society of Chemistry Emerging Technology Competition and our work has inspired others to build their own variants of the reactor. We are currently working with a consortium of large pharmaceutical companies to exploit commercially our reactor technology for manufacturing. The Covid pandemic occurred during Photo-Electro and the flexibility of the Programme Grant has allowed us to adapt our research to address some of the manufacturing challenges revealed by the pandemic, in particular the possibility of using continuous photochemistry and electrochemistry for more cost-effective manufacture of generic drugs so that their production could be re-shored to the UK. Clearly, the pandemic was disruptive for our PDRAs and the PhD students associated with Photo-Electro. Happily all of them have completed their work and found employment in jobs nearly all of which exploit technical skills acquired working on Photo-Electro. During the time of Photo-Electro, one CI moved into industry and two more Cis took early retirement. These moves enabled us to bring two ECRs into Photo-Electro with positive effects both on the research and the ECRs' careers. The industrial potential of the pre-competitive research carried out during Photo-Electro is underlined by the growth in the number of our industrial stakeholders which rose to 26 by the end of the programme. Many of those partnerships have continued beyond the end of Photo-Electro and several have led to additional Innovate UK or direct funding for more competitive research for a particular stakeholder. Throughout Photo-Electro, we have had an active public outreach programme including an exhibit "Green Light on Chemistry" at the 2019 Royal Society Summer Exhibition which was attended by more than 10,000 members of the public, a presentation in 2018 to the All Party Parliamentary Group on Neglected Tropical Diseases at the Houses of Parliament, a presentation to the Office of the Chief Scientific Advisor of BEIS in 2019, and an on-line lecture to a Russian science festival (2019) which was viewed by >70k people in the first 24 hours, as well as several lectures to schools across the UK. |
